TL;DR: The outstanding efficacy and prolonged blood concentrations of OZ439 are the result of a design strategy that stabilizes the intrinsically unstable pharmacophoric peroxide bond, thereby reducing clearance yet maintaining the necessary Fe(II)-reactivity to elicit parasite death.

TL;DR: The SAR of ozonide carboxylic acid OZ78 was described as the first part of the search for a trematocidal synthetic peroxide drug development candidate and it was found that relatively small structural changes to 1 resulted most commonly in loss of activity against Fasciola hepatica in vivo.
