Journal ArticleDOI
Comparative Performance of SARS-CoV-2 Detection Assays Using Seven Different Primer-Probe Sets and One Assay Kit.
Arun K. Nalla,Amanda M. Casto,Amanda M. Casto,Meei Li W. Huang,Garrett A. Perchetti,Reigran Sampoleo,Lasata Shrestha,Yulun Wei,Haiying Zhu,Keith R. Jerome,Keith R. Jerome,Alexander L. Greninger +11 more
TL;DR: All assays tested were found to be highly specific for SARS-CoV-2, with no cross-reactivity with other respiratory viruses observed in the authors' analyses regardless of the primer-probe set or kit used.
Journal ArticleDOI
Direct RT-qPCR detection of SARS-CoV-2 RNA from patient nasopharyngeal swabs without an RNA extraction step
Emily A. Bruce,Meei Li Huang,Garrett A. Perchetti,Scott Tighe,Pheobe Laaguiby,Jessica J Hoffman,Diana L Gerrard,Arun K. Nalla,Yulun Wei,Alexander L. Greninger,Alexander L. Greninger,Sean A. Diehl,David J. Shirley,Debra G.B. Leonard,Christopher D. Huston,Christopher D. Huston,Beth D. Kirkpatrick,Beth D. Kirkpatrick,Julie A. Dragon,Jessica W. Crothers,Keith R. Jerome,Keith R. Jerome,Jason Botten +22 more
TL;DR: Whether SARS-CoV-2 RNA could be detected from NP samples via a direct RT-qPCR assay that omits the RNA extraction step altogether is determined and has the potential to ease supply choke points to substantially expand COVID-19 testing and screening capacity.
Posted ContentDOI
DIRECT RT-qPCR DETECTION OF SARS-CoV-2 RNA FROM PATIENT NASOPHARYNGEAL SWABS WITHOUT AN RNA EXTRACTION STEP.
Emily A. Bruce,Meei Li Huang,Garrett A. Perchetti,Scott Tighe,Pheobe Laaguiby,Jessica J Hoffman,Diana L Gerrard,Arun K. Nalla,Yulun Wei,Alexander L. Greninger,Alexander L. Greninger,Sean A. Diehl,David J. Shirley,Debra G.B. Leonard,Christopher D. Huston,Christopher D. Huston,Beth D. Kirkpatrick,Beth D. Kirkpatrick,Julie A. Dragon,Jessica W. Crothers,Keith R. Jerome,Keith R. Jerome,Jason Botten +22 more
TL;DR: Direct RT-qPCR could be a front-line approach to identify the substantial majority of COVID-19 patients, reserving a repeat test with RNA extraction for those individuals with high suspicion of infection but an initial negative result.
Posted ContentDOI
RT-qPCR DETECTION OF SARS-CoV-2 RNA FROM PATIENT NASOPHARYNGEAL SWAB USING QIAGEN RNEASY KITS OR DIRECTLY VIA OMISSION OF AN RNA EXTRACTION STEP
Emily A. Bruce,Meei-Li Huang,Garrett A Perchetti,Scott Tighe,Jessica J Hoffman,Pheobe Laaguiby,Diana L Gerrard,Arun K. Nalla,Arun K. Nalla,Yulun Wei,Yulun Wei,Alexander L Greninger,Sean A. Diehl,David J. Shirley,Debra G.B. Leonard,Christopher D. Huston,Christopher D. Huston,Beth D. Kirkpatrick,Beth D. Kirkpatrick,Julie A. Dragon,Jessica W. Crothers,Keith R. Jerome,Jason Botten +22 more
TL;DR: It is reported here that the RT-qPCR assay can be performed directly on patient sample material from a nasal swab, without the need for an RNA extraction step of any kind, and should be applicable throughout the world.
Journal ArticleDOI
Validation of SARS-CoV-2 detection across multiple specimen types.
Garrett A. Perchetti,Arun K. Nalla,Meei-Li Huang,Haiying Zhu,Yulun Wei,Larry Stensland,Michelle A. Loprieno,Keith R. Jerome,Alexander L. Greninger,Alexander L. Greninger +9 more
TL;DR: A modified CDC-based laboratory developed test is able to detect SARSCoV- 2 accurately with similar sensitivity across all sample types tested, and the N2 gene target was found to be most sensitive in CSF.
