Abstract: Hartmannella culbertsoni, a free-living soil ameba, undergoes differentiation to be transformed into a double-walled cyst when exposed to a nonnutrient medium containing magnesium ions and taurine (Raizada and Krishna Murti, 1971) . The transformation of the trophic form to mature cysts is usually preceded by a stage during induction when the ameba assumes a round and well-defined shape after the withdrawal of the pseudopodia. Evidence is adduced in this communication to show that cyclic 3', 5'-adenosine monophosphate (CAMP) can replace magnesium ions and taurine in the nonnutrient agar medium and that CAMP brings about a comparable degree of encystation of the ameba. The presence of magnesium ions and taurine has also been shown to result in a threeto fourfold increase in the rate of synthesis of CAMP by the ameba .

Abstract: Hartmannella culbertsoni trophozoites are transformed into viable cysts on exposure to a non-nutrient agar medium containing 15 mM MgCl 2 and 20 mM taurine. Amebae differentiating in this encystment medium incorporate more uracil-2- 14 C into RNA and more leucine-1- 14 C or valine-1- 14 C into proteins than controls. Encysting organisms incorporate significantly more glucose-U- 14 C into cellulose and glucosamine-1- 14 C into mucopolysaccharides. Incorporation of glucose-U- 14 C into cellulose and of glucosamine-1- 14 C into mucopolysaccharides are inhibited by actinomycin D or cycloheximide.

Abstract: SYNOPSIS. Excystment of Schizopyrenus russelli was associated with increase in DNA, RNA, and protein. Both excystment and these macromolecular increases were sensitive to ultraviolet radiation. The effect of UV was partly reversed by incubation in the dark at 28 C, but not at 4 C. Excystment from cysts preincubated in the excystment medium (before UV treatment) was relatively resistant to radiation.

Abstract: 1. The effects of intracerebroventricular or intrathecal administration of glycine on some somatic reflexes in chloralosed cats were investigated. 2. Glycine produced marked inhibition of the flexor reflex (FR) and crossed extensor reflex (CER) at low doses (2·5-5·0 mg) but even large doses (up to 40·0 mg) had no effect on the facilitation or inhibition of the patellar reflex produced by stimulation of either the reticular formation or the sciatic nerve. The linguomandibular reflex (LMR) could be blocked by 10-20 mg glycine. 3. Glycine antagonized strychnine-induced facilitation of FR, CER and LMR and was 5-6 times as active as γ-aminobutyric acid in this respect. 4. The possibility of glycine being a transmitter in spinal pathways is discussed.

Abstract: SYNOPSIS. Serologic reactions elicited by sonically ruptured trophic and cystic forms of Hartmannella culbertsoni were studied. The antigens of trophic amoebae reacted with their homologous rabbit antiserum showing multiple precipitin lines which could not be seen when the reacting antigens were treated with trypsin prior to application on the Ouchterlony plates. Antigens of trophic amoeba did not react with antiserum against cysts. Cyst antigens reacted with their homologous antiserum only after trypsin treatment. Antigens prepared from trophozoites excysting from cysts reacted positively with the antiserum against antigens of trophic amoebae. Antigens of trophic as well as cystic forms fixed guinea pig complement in presence of their homologous antisera. With the trophic form, this property was abolished after trypsin treatment. Non-specific complement fixation mediated by cyst antigens was abolished by treatment with cellulase. Antiserum against trophic amoebae immobilized trophozoites and, in the presence of guinea pig complement, led to their lysis.

Abstract: 0730 09 1. l-Threonine dehydratase (E.C. 4.2.1.16) activity in the cell-free extracts of the trophozoites of Hartmannella culbertsoni has been purified 20-fold. 2. The enzyme requires pyridoxal-5-phosphate for its activity and is not cold-labile. Kinetic studies have shown that there is homotropic cooperative interaction between the substrate molecules. The enzyme is susceptible to end-product regulation, l-isoleucine being a feed back inhibitor and l-valine can partially reactivate the l-isoleucine inhibited enzyme. Some nucleotides stimulate the activity of the enzyme indicating allosteric nature of the enzyme. 3. Certain amoebicidal drugs and antibiotics have marked inhibitory effect on the enzyme.
