Abstract: A Gram-staining-negative, non-motile, spore-forming, rod-shaped, marine bacterial strain, CL-KR2T, was isolated from tropical seawater near Kosrae, an island in the Federated States of Micronesia. Analysis of the 16S rRNA gene sequence of strain CL-KR2T revealed a clear affiliation with the genus Gracilimonas . Based on phylogenetic analysis, strain CL-KR2T showed the closest phylogenetic relationship to Gracilimonas tropica CL-CB462T, with 16S rRNA gene sequence similarity of 96.6 %. DNA–DNA relatedness between strain CL-KR2T and G. tropica CL-CB462T was 6.7 % (reciprocal 9.5 %). Strain CL-KR2T grew in the presence of 1–20 % sea salts and the optimal salt concentration was 3.5–5 %. The temperature and pH optima for growth were 35 °C and pH 7.5. The major cellular fatty acids (≥10.0 %) of strain CL-KR2T were iso-C15 : 0, summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7c) and iso-C17 : 1ω9c and the only isoprenoid quinone was MK-7. The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, an unidentified phospholipid, two unidentified glycolipids and two unidentified lipids. The genomic DNA G+C content of strain CL-KR2T was 43.2 mol%. The combined phenotypic, chemotaxonomic and phylogenetic data showed that strain CL-KR2T could be distinguished from the only member of the genus Gracilimonas with a validly published name. Thus, strain CL-KR2T should be assigned to a novel species in the genus Gracilimonas , for which the name Gracilimonas rosea sp. nov. is proposed. The type strain is CL-KR2T ( = KCCM 90206T = JCM 18898T).

Abstract: A novel cream-pigmented marine bacterium, designated strain YJ-T1-11T, was isolated from a tidal flat at Yeongjong-do, Republic of Korea. Cells were rod-shaped, non-motile, aerobic, Gram-reaction-negative, oxidase-positive and catalase-positive. Phylogenetic analysis of 16S rRNA gene sequences indicated that strain YJ-T1-11T clustered with Gemmobacter fontiphilus JS43T (98.3 %) within the genus Gemmobacter and its closest neighbours were G.emmobacter aquatilis DSM 3857T (98.5 %), Gemmobacter aquaticus A1-9T (98.4 %), G emmobacter lanyuensis Orc-4T (98.4 %), G emmobacter fontiphilus JS43T (98.3 %), G emmobacter caeni DCA-1T (98.2 %), G emmobacter nanjingensis Y12T (97.5 %) and G emmobacter tilapiae Ruye-53T (97.2 %). Average nucleotide identity values between the genome sequences of strain YJ-T1-11T and the related type strains ranged from 77.08 to 90.48 %. The predominant fatty acid of strain YJ-T1-11T was summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c). The major isoprenoid quinone was Q-10 and the DNA G+C content was 65.6 mol%. The polar lipid profile consisted of phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, diphosphatidylglycerol and three unidentified lipids. The DNA–DNA relatedness values between strain YJ-T1-11T and the type strains of the 12 phylogenetically related species of the genus Gemmobacter were 23.6–53.7 %. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain YJ-T1-11T is considered to represent a novel species of the genus Gemmobacter , for which the name Gemmobacter lutimaris sp. nov. is proposed. The type strain is YJ-T1-11T (=KCTC 62715T=JCM 32828T).

Abstract: Plant viruses continue to cause diseases on economically important crops. Therefore, numerous attempts to produce virus resistant plants have been reported by using the mechanisms such as host mediated protection and virus mediated protection. Here, a novel strategy of targeting viral RNA itself, rather than viral gene products, is presented to generate virus-resistant transgenic plants. A catalytic single chain variable antibody, 3D8 scFv, which has RNase activities, was functionally expressed in the cytosol of Nicotiana tabacum. We found that progenies of the transgenic tobacco plant acquired complete resistances against four ss-RNA tobamoviruses and one cucumovirus tested without viral accumulation and delayed onset of disease symptoms. The results showed that the resistance observed in 3D8 scFv transgenic plants was caused by the RNase activity of 3D8 scFv itself, not by RNA-mediated gene silencing mechanism. Taken together, we suggested that newly gained resistance of the 3D8 scFv transgenic plants to five ss-RNA viruses most likely resulted from the RNase activity of 3D8 scFv.

Abstract: [1] Eleven climate models, one high-resolution and ten low-resolution, were analyzed to investigate the response of the northwestern Pacific under a global warming scenario. Application of scenario A1B of the Special Report on Emission Scenarios (SRES) weakens (intensifies) the southern (northern) part of the interior subtropical gyre both in high-resolution and low-resolution model. Such a dipole type change is mainly due to a basin-scale dynamic atmosphere-to-ocean process. Namely, under global warming the Hadley circulation is weakened and expanded poleward. The Ferrel circulation is also displaced poleward, leading to weakening of ascending (descending) air motion and a high (low) sea level pressure anomaly in the northwestern (southeastern extratropical) North Pacific. Finally, a negative wind stress curl anomaly developed along the zero wind stress curl line of the present-day climate to enhance the northern part of the gyre. The high-resolution model results show greater changes in the structure of the Kuroshio and Kuroshio Extension, with strong intensification of the Kuroshio Extension front and jet, while in the low-resolution models the changes are small. The Kuroshio between Taiwan and the southern coast of Japan is significantly intensified in the high-resolution model results, but is slightly weakened in the ensemble of the low-resolution models.
