Abstract: Objective To evaluate the effects of electric-acupuncture and fluoxetine on depression and neurological function of post-stroke depression(PSD) patientsMethods A total of 95 patients with PSD were randomly divided into the group A,B and C,they were treated with electric-acupuncture,medicine,and electric-acupuncture combined with medicine respectivelyThe degree of depression and neurological function defect,side-effects were evaluated by hamilton depression scale(HAMD),the clinical neurological function defect scale and the treatment emergent symptom scale(TESS) before and after treatmentResults After treatment for 6 weeks,HAMD scores,the clinical neurological function defect scale and TESS scores were significantly different in 3 groups(P005),which in the group C was much better than those in group A and B(P005)Conclusion Electric-acupuncture combined with fluoxetine could regulate depression and improve neurological function and reduce side effects of medicine in treating PSD patientsIt is deserved to be applied to clinical use

Abstract: Objective To investigate the association between the polymorphism of S2 locus allele in ADAM33 gene and COPD.Methods Allele-specific polymerase chain reaction(AS-PCR) and DNA sequencing were used to determine the polymorphism of S2 locus allele in ADAM33 gene in 112 patients with COPD and 105 healthy controls.Results The genotype distributions of S2 locus allele in ADAM33 gene in case group and control group did not deviate from Hardy-Weinberg equilibrium.The frequencies of the genotypes(CC,CG,GG) were 54.5%,34.8% and 10.7%;in the 112 COPD patients and 69.5%,26.7% and 3.8% in the 105 controls,respectively,showing a significant difference in the distribution of the genotypes(CC,CG,GG) between COPD patients and healthy controls(χ2=6.662,P0.05).The frequencies of the alleles(C,G) were 0.719 and 0.281 in the COPD patients and 0.829 and 0.171 in the controls,respectively,also showing a significant difference in the allele frequency between them(χ2=7.424,P0.05).Conclusion The polymorphism of S2 locus allele in ADAM33 gene is associated with the susceptibility to COPD.

Abstract: Objective To observe the single nucleotide polymorphisms(SNPs) located at 607 and 137 in the promoter region of serum interleukin-18(IL-18) gene in acute cerebral infarction(ACI)patients.Methods Polymerase chain reaction with sequence specific primer(PCR-SSP) was used to genotype the SNPs of 607C/A and 137G/C in 98 ACI patients(observe group)and 100 healthy controls(control group).Results There were no significant difference in the distribution of genotypic frequencies and allelic frequency at position 607.The frequencies of 137 GG genotype was significantly higher in ACI patients(P0.05),while 137 GC was significantly decreased in ACI patients compared to the control groups(P0.05).Significant difference was also found in allelic frequency at position 137 between ACI patients and control(P0.05).Conclusion The IL-18 promoter gene polymorphism at position 607 is not associated with ACI,but the polymorphism at position 137 is associated with ACI and C allele may be protected against ACI.

Abstract: Objective To observe the morphological and functional changes of the proprioceptors in the partially injuried ACL and explore the mechanism.Methods The ACL in 12 rabbits was damaged to make animal model.At 2-,4-and 6-month post-operation,the ACL in 4 rabbits separately was examined with electrophysiologic study(including SEPs and EMG) and gold chlorid staining method to judge the morphological and functional changes of the neuromechanism in the ligament tissue.Results The incubation period and wave amplitude of the SEPs and EMG degraded gradually following time.The quantity of the proprioceptors in the injuried ACL gradually reduced following time,and the morphogenesis of the proprioceptors changed,including the volume of proprioceptors diminuted and part structure depleted.Conclusion The proprioceptors in the ligament can still have some function after ACL being injuried,but the effect weakened gradually following time.The mechanism might be that the partially injuried ACL loose gradually due to its decreased strength,and thus the proprioceptors in the ligament can not be stimulated by normal stress produced in the course of knee motion.

Abstract: Objective To investigate the expression of cyclooxygenase 2(COX-2) and vascular endothelial growth factor(VEGF) in osteosarcoma and explore the significance.Methods 45 patients with osteosarcoma were treated with amputation or prosthesis replacement.Osteosarcoma and corresponding adjacent(5.0 cm at the outer edge of the tumor tissue) tissue samples were obtained during operation.The expression of COX-2,VEGF and MVD marked by CD34 of osteosarcoma and their adjacent tissues were detected by immuno histochemistry.Results The positive rates of COX-2,VEGF and MVD in osteosarcoma tissues were 80.0%,75.6% and(43.6±11.4)stripe/HP,respectively,which were higher than those in adjacent tissues(P all 0.05).In patients with osteosarcoma,the expression of COX-2,VEGF and MVD were associated with Enneking stage(P all 0.05).There was a positive relationship between the expression of COX-2 and VEGF,the expression of COX-2 and VEGF were positively correlated with MVD(r=0.75,0.58,respectively,P all 0.05).The MVD values in both COX-2 and VEGF positive samples were higher than those in COX-2 negative and VEGF negative samples(P0.05).Conclusions COX-2 and VEGF are highly expressed in osteosarcoma and expression of them are associated with surgical stage of osteosarcoma,may involved in the occurrence and development of osteosarcoma,the mechanism maybe contributing to angiogenesis in osteosarcoma.

Abstract: Objective To investigate the effects of 18β-glycyrrhetinic acid(18β-GA)on the proliferation,adhesion,invasion and the expression of FAK,MMP-9 of human highly metastatic ovarian carcinoma HO-8910PM cells.Methods HO-8910PM cells were treated with 18β-GA,then MTT assay was used to determine the growth inhibition;Transwell chamber assay was performed to determine the effect on invasion of the cells by 18β-GA;cell adhesion assay was performed to detect the adhesion of HO-8910PM;Western blot analysis was performed to detect the protein expreesion of FAK and MMP-9.Results 18β-GA inhibited the growth,proliferation,adhesion,invasion in cells;18β-GA also down-regulated the expression of FAK,MMP-9 protein.Conclusion 18β-GA can inhibit the proliferation,adhesion and invasion of HO-8910PM cells,the mechanism may be involved in the reduction of the expression level of FAK,MMP-9.

Abstract: Objective To investigate the relatioship between the prostate stem cell antigen(PSCA gene) polymorphisms and susceptibility to Tibetan gastric cancer in Qinghai area.Methods Genotypes were determined by PCR-based denaturing high-performance liquid chromatography analysis and direct DNA sequencing in 60 incident cases with gastric cancer(gastric cancer group) and 60 controls recruited(control group)in Qinghai area.Results The frequency of G/A in gastric cancer group was significantly higher than that in the control group(P0.05);no significant difference for the frequency of A/A and G/G was found between the two groups.Conclusion The polymorphism of PSCA gene rs2976392 are associated with the susceptibility of Tibetan gastric cancer in Qinghai area;people with PSCA G/A genetype is susceptible to gastric cancer.

Abstract: Objective To detect the expression of ADAMTS-4(a disintegrin and metalloproteinase with thrombospondin motifs) in metastasis and invasion of breast tumor.Methods The expression of ADAMTS-4 and ARGxx(one of ADAMTS-4 degradate aggrecan products),were detected in 20 cases of breast cancer,22 cases of breast fibroadenoma by immunohistochemical method.Results ADAMTS-4 and ARGxx were detectable in 80% of breast cancer,which was significantly higher than those in breast fibroadenoma 27% and 18% respectively.The optical density of ADAMTS-4 and ARGxx in breast cancer were 0.235 8±0.059 5 and 0.170 0±0.051 0,they were also significantly higher than those of breast fibroadenoma,which were 0.215 0±0.045 5 and 0.134 2±0.036 3.Conclution ADAMTS-4 is overexpressed in breast cancer,and its activity is also enhanced.The enzyme is involved in tumor metastasis and invasion by degrading aggrecan and thus changes cell adhesion and communication.

Abstract: Objective To investigate the effects of peroxisome proliferator-activated receptor α agonist fenofibrate combined with cisplatin on proliferation and apoptosis of human lung cancer A549 cell line and the possible mechanism.Methods A549 cells were exposed to fenofibrate,cisplatin and fenofibrate combined with cisplatin respectively.MTT method was used to investigate proliferation.Hoechest stain was applied to analyze the apoptosis effects.Flow cytometry was used to detect the apoptosis.A549 cells caspase-3 and survivin mRNA expression changes after treatments were detected by RT-PCR.Results Fenofibrate could inhibit the proliferation of A549 cells in dose-time relationship.Compared with either fenofibrate or cisplatin alone,combining fenofibrate with cisplatin increased the growth inhibition rate of A549 cells(P0.05).More apoptotic cells were detected in combined treatment group than separate treatment group by Hoechest stain and flow cytometry.RT-PCR results showed the expression of caspase-3 mRNA was up regulation,then the expression of survivin mRNA was down regulation.Conclusion Combining fenofibrate with cisplatin can enhance the role of cisplatin killing lung cancer A549 cells,which may be a result of up-regulating caspase-3 and down-regulating survivin.

Abstract: Objective To investigate the effects of phospho-Akt(p-Akt),phosphatase and tensin homolog deleted on chromosome ten(PTEN) and P-glycoprotein(P-gp)in the occurrence and development of breast cancer.Methods A series of 81 primary axillary lymph node negative(LNN) breast cancer specimens were performed by immunohistochemical staining to detect the expression of p-Akt,PTEN and P-gp.The correlation between the three indexes and clinicopathological factors of breast cancer were analyzed by χ2-test,exact propability and Cox ′s proportional hazard model.Results A marked increased activity of p-Akt was detected in breast cancer,which was significantly higher than that in normal breast tissues(P=0.034).The PTEN expression was 63.0%,which was significantly lower than that in normal tissues(P=0.001).The overexpression of p-Akt and loss of PTEN was related with tumor differentiation,ER negative,recurrence,metastasis and 5-years survival rates.The positive rate of P-gp was 39.5%,related with recurrence and distant metastasis.Multivariate analysis found that p-Akt expression was also signigicantly positively correlated with P-gp expression(r=0.548,P=0.045),and also expression of p-Akt(P=0.04) and PTEN(P=0.02) were independent prognostic factors for prognosis of LNN breast cancer.Conclusions P-Akt,PTEN and P-gp may play certain effects in the occurrence and development of breast cancer,to detect the level of the three indexes is helpful for the prognosis evaluation.

Abstract: Objective To investigate the effects of ginsenoside Rg3 on cell proliferation of breast cancer cell line MDA-MB-231 and the possible mechanism involved.Methods MDA-MB-231 cells at logarithmic growth phase were obtained,and were cultured with ginsenoside Rg3 of different concentrations(50、100、200、400、600 μmol/L).Cells cultured without ginsenoside Rg3 were served as controls.The proliferation of cells were detected by MTT assay,and the expression of Cyclin E and CDC25A was examined by Western blot,respectively.Results The growth rates on MDA-MB-231 cells treated with ginsenoside Rg3 of low concentration(≤200 μmol/L)increased,but those treated with ginsenoside Rg3 of high concentration(≥400 μmol/L)growed more slowly,compared with control group.The expression of cellular Cyclin E and CDC25A rose when exposed to ginsenoside Rg3 of low concentration(≤200 μmol/L)while declined when exposed to ginsenoside Rg3 of high concentration(≥400 μmol/L).Conclusions Ginsenoside Rg3 of high concentration(≥400 μmol/L)can inhibit the growth of MDA-MB-231.This effect may involve its suppression of cell-cycle related protein Cyclin E and CDC25A.

Abstract: Objective To investigate NFATC1 gene rs754093 polymorphism associated with hypertensive cerebral hemorrhage in Han population of China.Methods The subject of observe group were consecutively recruited from northern China region patients with a total of 230 cases.The controls were health people with a total of 233 cases.The snp rs754093 of NFATC1 was genotyped by PCR-RFLP.Results THE observe group T / T、 T / G 、G/G genotype frequencies and T、G allele frequencies were 34.1%、48.3%、17.4%、58.5%、41.5% separately,the controls group were 44.2%、44.6%、11.2%、66.5%、33.5% separately.There was significant difference between two groups(P0.05).Comparing with TT genotype,the GG+TG genotype could increase 51.4% of the risk of cerebral hemorrhage.Compare with T allele the G allele can increase 41.1% of the risk of cerebral hemorrhage,.Conclusions NFATC1 gene rs754093 polymorphism is associated with hypertensive cerebral hemorrhage in Han population of China.The G / G genotype is the independent risk factor of cerebral hemorrhage in Han population of China.

Abstract: Objective To study the early diagnosis value of serum cystatin C and urine microalbumin on renal injures of gestational diabetes mellitus patients.Methods The concentration of cystatin C and urine microalbuminuria were determined by nephelometry method.The levels of the blood urea nitrogen(BUN) and the creatinine(Scr) were measured by enzymatic method.Results The levels of cystatin C,urine microalbuminuria and Scr in patients with gestational diabetes mellitus were obviously higher than those of the normal group(P0.05 or0.01).The level of BUN had no significant difference in the two group(P0.05).Conclusion The cystatin C and urine microalbuminuria quantified together have important significance in the early diagnosis in patients with gestational diabetes mellitus.

Abstract: ObjectiveTo investigate the anti-tumor effects of total polysaccharide(TPS) extracted from Shiquan Dabu Decoction in vitro,so as to provide basis for its clinical application and research of anti-tumor drugs.MethodsThe human lung cancer cells A549,hepatoma cells Bel-7402,colorectal carcinoma cells HCT-8 and cervical carcinoma cells Hela cultured in vitro were treated with TPS of 200,100,50 μg/ml,the inhibitory rate of proliferation were measured by MTT assay at 24,48,72 h after the treatment.ResultsIR reached the highest in the A549 cells when treated with TPS of 200 μg/ml for 48 hs;IR in the other three kinds of cells increased gradually with the increasing of TPS concentration and prolonging of treating time,reached the highest at 200 μg/ml for 72 hs,which showed dose and time-dependent manner.ConclusionsTPS could inhibit the growth of tumor in vitro;and hope to be used for the treatment of tumor as anti-tumor drugs.

Abstract: ObjectiveTo express the structural protein VP1 of enterovirus 71 in E.coli,and identify its antigenicity initially.MethodsThe target gene was amplified by PCR and cloned into expression vector pET30a(+).The recombinant plasmid was transformated into E.coli Transetta(DE3),the expression and antigenicity of recombinant protein was analyzed initially by SDS-PAGE and Western Blot assays;the best expression condition of target protein was investigated.ResultsThe fusion protein about 30 kD was obtained,and Western Blot assays showed it had good antigenicity;the best expression condition was 37 ℃,1.0 mmol/L IPTG and 4 h.ConclusionsEV71 VP1 antigen can be expressed and identified;this study provides foundation for the vaccinal research and manufacture the detection kit of EV71.

Abstract: Objective To study the effects of p38 mitogen on cAMP response element binding protein(CREB) expression in microglial cells treated with nutmeg ethanol extract.Methods WST-8 kit was used to detect the viability of BV2 murine microglial cells treated with the nutmeg ethanol extract.The effects of lipopolysaccharide(LPS) and nutmeg ethanol extract on the expression of p38MAPK,p-p38MAPK and CREB were measured by Western blot technology.Result Nutmeg ethanol extract did not affect cell viability,but could inhibit the expression of p-p38MAPK and CREB induced by LPS.Conclusion Nutmeg ethanol extract could inhibit the p-p38MAPK and CREB proteins expression induced by LPS.

Abstract: Objective To explore the morphological features,diagnosis and differential diagnosis of the fetal cardiac rhabdomyomas.Methods Four cases of fetal cardiac rhabdomyomas by autopsy were reperted.The tissue sections were stained with HE,immunohistochemistry and PAS.Results Fetal cardiac rhabdomyomas was single or multiple neoplasm with clear margin.Histologically,the "spider cells" were typical.The tumor cells expressed myoglobin and desmin.PAS result was positive.Conclusion Fetal cardiac rhabdomyomas is the most common cardiac tumor in fetus,which can be single or multiple.

Abstract: Objective To observe the changes of apoptosis in NIT-1 cell induced by high glucose in the Raman spectra of NIT-1 insulin beta cells,and explore a novel approach for studying the apoptosis cells.Methods According to different concentrations of glucose and different culture time,the cells were divided into 6 groups.The apoptosis rate was measured by the flow cytometry,and the spectra was measured by the laser tweezers-Raman spectroscopy system.The differences in the apoptosis rate were compared and in the spectra information was analyzed in the 6 groups.Results The apoptosis rate of NIT-1 cell increased significantly in high glucose cultured medium,compared to control medium,and the apoptosis rate increased with the represented of induction time(P0.05).Most of the floating cells were in a state of late apoptosis after cultured.The Raman spectra showed that the area of peaks at 1 001/cm and 1 337/cm which the contents of protein decreased significantly with the extension of induction time(P0.05).Conclusions High glucose can induce NIT-1 cell apoptosis,and the apoptosis rate increases with the extension of induce time.Proteins and nucleic acid are less in apoptotic cells than in normal cells which can be reflected by Raman spectroscopy.

Abstract: Objective To explore the correlation between susceptibility to inflammatory bowel disease(IBD)in Chinese zhuang population of Guangxi province and ATG16L1 gene single nucleotide polymorphisms(SNP),rs2241880.Methods The Intestinal tissue of 146 patients of IBD and 164 healthy people were collected.Genomic DNA was extracted from intestinal tissueusing with phenol chloroform methodology.Polymerase chain reaction was used to amplify the ATG16L1 gene,and products were sequenced using the sequencer.The sequenced results were compared to the normal sequence in Genbank.Results No significant differences were noted in the ATG16L1 gene SNP site rs2241880 polymorphisms among the patients with Crohn' s disease,ulcerative colitis and the control subjects(P0.05).Conclusions The SNP rs2241880 of ATG16L1 are not found to increase the susceptibility of IBD in Chinese zhuang population of Guangxi province.

Abstract: Objective To study the preventive and therapeutic effect of tanshinol on bleocin-induced pulmonary fibrosis in rats and possible mechanism.Methods Forty male rats were assigned randomly into four groups: normal group,model group,tanshinol group,dexamethasone group with 10 rats in each group.The pulmonary fibrosis rat models were founded by infusing 5 mg/kg bleocin into trachea.All rats were killed when feeding for 28 days.The SOD,MDA and HYP contents in lung tissue were assayed in order to observe the influence of Tanshinol on pulmonary fibrosis.Results SOD activity of lung tissue was significantly higher,and lower MDA,HYP contents in tanshinol group than that in the normal group.There was no statistical difference between tanshinol group and dexamethasone group.Conclusion Tanshinol can relieve and inhibit pulmonary fibrosis of rats induced by bleocin.

Abstract: Objective To study the prognosis of atherothrombosis ischemic stroke patients with antiplatelet,lipid-lowering or antioxidants therapy in the acute phase.Methods A total of 689 atherothrombosis ischemic stroke patients were divided into controlled groups and observed groups based on whether patients accepted antiplatelet,statins or probucol antioxidant therapy.The improvement rate,recurrence rate,the incidence of unfavorable prognosis and mortality rate were evaluated by modified Rankin scale(mRs) after 12 months follow-up.Results After three months and twelve months follow-up,the improvement rate was higher in observed groups,but the incidence of unfavorable prognosis and mortality rate were lower in observed groups than those in controlled groups(P0.05),however there were no statistically difference in recurrence rate between observed groups and controlled groups(P0.05).Conclusions Antiplatelet,statins lipid-lowering or probucol antioxidant therapy in the acute phase can not only improve the prognosis of atherothrombosis ischemic stroke patients,but also decrease the incidence of unfavorable prognosis and mortality rate.

Abstract: Objective To investigate the effects of total phenolics from abnormal savda munziq combined with cisplatin or docetaxel on HeLa cell line of cervical cancer cultured in vitro.Methods HeLa cells were treated with different drug concentrations: total phenolics combined with cisplatin and total phenolics combined with docetaxel,we chose MTT assay to test the inhibiting effects and Flow cytometry to examine cell apoptosis rate.Microscope was used to observe the growth of HeLa cells.Results The inhibiting effect and apoptosis rate of total phenolics combined with cisplatin was greater than cisplatin(P0.05),this phenomenon also appeared in total phenolics combined with docetaxel.Conclusion Total phenolics from abnormal savda munziq combined with cisplatin or docetaxel can inhibit the growth of HeLa cells obviously,and induce apoptosis in vitro,the combinations have synergistic actions.
