TL;DR: The unstable metabolic intermediate 1Δ-pyrroline-5-car☐ylic acid (P5C or glutamic semialdehyde) can be isolated as a chemically pure solid by preparing it in dilute solution by periodate oxidation of dl-hydroxylysine and isolating it as a pure 2,4-dinitrophenylhydrazone from which P5C can be quantitatively regenerated.

TL;DR: The quantitative determination of the pyridine nucleotides and of substrates which can be brought into stoichiometric reaction with them has been hampered by the lack of reliable extinction coefficients for these substances.
