A CRISPR Dropout Screen Identifies Genetic Vulnerabilities and Therapeutic Targets in Acute Myeloid Leukemia
Konstantinos Tzelepis,Hiroko Koike-Yusa,Etienne De Braekeleer,Yang Li,Emmanouil Metzakopian,Oliver M. Dovey,Annalisa Mupo,Vera V. Grinkevich,Meng Li,Milena Mazan,Malgorzata Gozdecka,Shuhei Ohnishi,Jonathan L. Cooper,Miten Patel,Thomas McKerrell,Bin Chen,Ana Domingues,Paolo Gallipoli,Sarah A. Teichmann,Hannes Ponstingl,Ultan McDermott,Julio Saez-Rodriguez,Julio Saez-Rodriguez,Brian J. P. Huntly,Francesco Iorio,Cristina Pina,George S. Vassiliou,George S. Vassiliou,Kosuke Yusa +28 more
TL;DR: KAT2A inhibition demonstrated anti-AML activity by inducing myeloid differentiation and apoptosis, and suppressed the growth of primary human AMLs of diverse genotypes while sparing normal hemopoietic stem-progenitor cells.
Journal ArticleDOI
Mitochondria and ubiquitin-proteasomal system interplay: Relevance to Parkinson's disease
Ana Domingues,Daniela M. Arduíno,A. Raquel Esteves,Russell H. Swerdlow,Catarina R. Oliveira,Sandra M. Cardoso +5 more
TL;DR: The results suggest that a mitochondrial alteration leads to an imbalance in the cellular oxidative status, inducing a proteasomal deregulation, which may exacerbate protein aggregation, and consequently degenerative events.
Journal ArticleDOI
WT1, MSH6 , GATA5 and PAX5 as epigenetic oral squamous cell carcinoma biomarkers - a short report
Ilda Patrícia Ribeiro,Francisco Caramelo,Francisco Batel Marques,Ana Domingues,Margarida Mesquita,Leonor Barroso,Hugo Prazeres,Maria José Julião,Isabel Poiares Baptista,Artur Ferreira,Joana B. Melo,Isabel M. Carreira +11 more
TL;DR: This is the first study that highlights this specific set of genes as epigenetic diagnostic and prognostic biomarkers in OSCC, and it is found that WT1 gene promoter methylation is a predictor of a better prognosis and that MSH6 and GATA5 gene promoters serve as predictors of a worse prognosis.
Journal ArticleDOI
Calcium-permeable α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors trigger neuronal nitric-oxide synthase activation to promote nerve cell death in an Src kinase-dependent fashion.
Renato Socodato,Felipe N. Santiago,Camila C. Portugal,Camila C. Portugal,Ana Domingues,Ana Raquel Santiago,João B. Relvas,António F. Ambrósio,Roberto Paes-de-Carvalho +8 more
TL;DR: The data presented herein highlight that Src modulation is a critical step in excitatory retinal cell death, and suggest NO pathway as a key regulatory signaling in AMPA-induced Src activation in neurons but not in glial cells.
Journal ArticleDOI
Towards a detailed anthropometric body characterization using the Microsoft Kinect.
Ana Domingues,Filipa Barbosa,Eduardo M. Pereira,Márcio Borgonovo dos Santos,Adérito Seixas,João Paulo Vilas-Boas,Joaquim Gabriel,Ricardo Vardasca,Ricardo Vardasca +8 more
TL;DR: A methodology is proposed and implemented for estimating anthropometric measures considering data provided by low-cost sensors, such as the Microsoft Kinect, and a more complete characterization of the whole body structure was achieved.
