Journal ArticleDOI
Structural basis for LIN54 recognition of CHR elements in cell cycle-regulated promoters
Aimee H. Marceau,Jessica Felthousen,Paul D. Goetsch,Audra N. Iness,Hsiau-Wei Lee,Sarvind Tripathi,Susan Strome,Larisa Litovchick,Seth M. Rubin +8 more
TL;DR: This work characterize the DNA-binding properties of LIN54 and describes the structural basis for recognition of a CHR sequence, and biochemically defines the CHR consensus as TTYRAA and determines that two tandem cysteine rich regions are required for high-affinity DNA association.
Journal ArticleDOI
The cell cycle regulatory DREAM complex is disrupted by high expression of oncogenic B-Myb.
Audra N. Iness,Jessica Felthousen,Varsha Ananthapadmanabhan,Fatmata Sesay,Siddharth Saini,Keelan Z. Guiley,Seth M. Rubin,Mikhail G. Dozmorov,Larisa Litovchick +8 more
TL;DR: It is shown that overexpression of B- myb disrupts the DREAM complex in human cells, and this activity depends on the intact MuvB-binding domain in B-Myb, and the rationale for targeting B-myb to restore cell cycle control is established.
Journal ArticleDOI
Structural mechanism of Myb-MuvB assembly.
Keelan Z. Guiley,Audra N. Iness,Siddharth Saini,Sarvind Tripathi,Joseph S. Lipsick,Larisa Litovchick,Seth M. Rubin +6 more
TL;DR: The structure and biochemical analysis provide an understanding of how oncogenic B-Myb is recruited to regulate genes required for cell-cycle progression, and the MMB interface presents a potential therapeutic target to inhibit cancer cell proliferation.
Journal ArticleDOI
Training the physician-scientist: views from program directors and aspiring young investigators
Christopher S. Williams,Audra N. Iness,Rebecca M. Baron,Olujimi A. Ajijola,Patrick J. Hu,Jatin M. Vyas,Robert A. Baiocchi,Alexander J. Adami,Jeremie M. Lever,Peter S. Klein,Linda L. Demer,Michael P. Madaio,Mark W. Geraci,Lawrence F. Brass,Melvin Blanchard,Robert A. Salata,Mone Zaidi +16 more
TL;DR: Consensus is shared from iterative workshops hosted by the Alliance of Academic Internal Medicine (AAIM) and the student-led American Physician Scientists Association (APSA) that standards for research training and tools for evaluating programmatic success are lacking.
Journal ArticleDOI
MuvB: A Key to Cell Cycle Control in Ovarian Cancer.
Audra N. Iness,Larisa Litovchick +1 more
TL;DR: The rationale for additional investigation of MuvB in high-grade serous ovarian cancer and the need for further translational research are highlighted.
