Journal ArticleDOI
The common PPARgamma Pro12Ala polymorphism is associated with decreased risk of type 2 diabetes.
David Altshuler,Joel N. Hirschhorn,Joel N. Hirschhorn,Joel N. Hirschhorn,Mia Klannemark,Cecilia M. Lindgren,Cecilia M. Lindgren,Marie-Claude Vohl,James Nemesh,Charles R. Lane,Stephen F. Schaffner,Stacey Bolk,Carl G. Brewer,Tiinamaija Tuomi,Tiinamaija Tuomi,Daniel Gaudet,Thomas J. Hudson,Thomas J. Hudson,Mark J. Daly,Leif Groop,Eric S. Lander +20 more
TL;DR: By analysing over 3,000 individuals, this work found a modest but significant increase in diabetes risk associated with the more common proline allele, which translates into a large population attributable risk—influencing as much as 25% of type 2 diabetes in the general population.
Journal ArticleDOI
Molecular scanning of the human PPARα gene: association of the L162V mutation with hyperapobetalipoproteinemia
Marie-Claude Vohl,Pierre Lepage,Daniel Gaudet,Carl G. Brewer,Christine Bétard,Patrice Perron,Ghislaine Houde,Christine Cellier,Janet Faith,Jean-Pierre Després,Kenneth Morgan,Kenneth Morgan,Thomas J. Hudson,Thomas J. Hudson,Thomas J. Hudson +14 more
TL;DR: Results suggest an association between the PPARalpha V162 allele and the atherogenic/hyperapolipoprotein B dyslipidemia among French Canadians.
Journal ArticleDOI
Mapping susceptibility genes for bipolar disorder: a pharmacogenetic approach based on excellent response to lithium.
Gustavo Turecki,Paul Grof,Eva Grof,D'Souza,Lebuis L,Marineau C,P. Cavazzoni,Anne Duffy,Christine Bétard,Petr Zvolský,Carrie Robertson,Carl G. Brewer,Thomas J. Hudson,Guy A. Rouleau,Martin Alda,Martin Alda +15 more
TL;DR: A complete genome scan using 378 markers spaced at an average distance of 10 cM in 31 families ascertained through excellent lithium responders provided original evidence suggesting that loci on 15q14 and 7q11.2 may be implicated in the pathogenesis of BD responsive to lithium.
Journal ArticleDOI
Identification of a chromosome 8p locus for early-onset coronary heart disease in a French Canadian population
James C. Engert,James C. Engert,Mathieu Lemire,Janet Faith,Diane Brisson,T. Mary Fujiwara,Nicole M. Roslin,Carl G. Brewer,Alexandre Montpetit,Corinne Darmond-Zwaig,Yannick Renaud,Carole Doré,Swneke D. Bailey,Andrei Verner,Gérald Tremblay,Julie St-Pierre,Christine Bétard,Jill Platko,Jill Platko,John D. Rioux,John D. Rioux,Kenneth Morgan,Thomas J. Hudson,Daniel Gaudet +23 more
TL;DR: Evidence is provided for a locus on chromosome 8 that contributes to CHD in a French Canadian population descended from a founder population that settled this region 300–400 years ago.
Journal ArticleDOI
Pedigree selection and tests of linkage in a Hutterite asthma pedigree.
Celia M. T. Greenwood,Alexandre Bureau,J. C. Loredo-Osti,Nicole M. Roslin,M. J. Crumley,Carl G. Brewer,T. M. Fujiwara,Darlene R. Goldstein,Kenneth Morgan +8 more
TL;DR: Using MCMC, the evidence for linkage in complex subpedigrees of several hundred individuals is evaluated, and hence, some of the co‐ancestry of this founder population is incorporated.
