Journal ArticleDOI
Everolimus improves the efficacy of dasatinib in PDGFRα-driven glioma.
Zachary Miklja,Viveka Nand Yadav,Rodrigo Cartaxo,Ruby Siada,Chase Thomas,Jessica R. Cummings,Brendan Mullan,Stefanie Stallard,Alyssa Paul,Amy K. Bruzek,Kyle Wierzbicki,Tao Yang,Taylor Garcia,Ian Wolfe,Marcia Leonard,Patricia L. Robertson,Hugh J. L. Garton,Daniel R. Wahl,Hemant Parmar,Jann N. Sarkaria,Cassie Kline,Sabine Mueller,Theodore Nicolaides,Chana Glasser,Sarah Leary,Sriram Venneti,Chandan Kumar-Sinha,Arul M. Chinnaiyan,Rajen Mody,Manjunath P. Pai,Timothy N. Phoenix,Bernard L. Marini,Carl Koschmann +32 more
TL;DR: Efficacy of dasatinib treatment of PDGFRα-driven HGG is improved with everolimus and suggests a promising route for improving targeted therapy for this patient population.
Journal ArticleDOI
Serial H3K27M cell-free tumor DNA (cf-tDNA) tracking predicts ONC201 treatment response and progression in diffuse midline glioma
Evan Cantor,Kyle Wierzbicki,Rohinton Tarapore,Karthik Ravi,Chase Thomas,Rodrigo Cartaxo,Viveka Nand Yadav,Ramya Ravindran,Amy K Bruzek,Jack Wadden,Vishal John,Clarissa Babila,Jessica R. Cummings,Abed Rahman Kawakibi,Sunjong Ji,Johanna Ramos,Alyssa Paul,Dustin Walling,Marcia Leonard,Patricia L. Robertson,Andrea Franson,Rajen Mody,Hugh J. L. Garton,Sriram Venetti,Yazmin Odia,Cassie Kline,Nicholas A Vitanza,Soumen Khatua,Sabine Mueller,Joshua E. Allen,Sharon Gardner,Carl Koschmann +31 more
TL;DR: Clinical utility in terms of predicting progression and sustained response and possible differentiation of pseudo-progression and pseudo-response is demonstrated and the feasibility and potential utility of serial cf-tDNA in both plasma and CSF of DMG patients to supplement radiographic monitoring is demonstrated.
Journal ArticleDOI
Therapeutic targeting of prenatal pontine ID1 signaling in diffuse midline glioma
Dana Messinger,Micah Harris,Jessica R. Cummings,Chase Thomas,Tao Yang,Stefan R. Sweha,Rinette Woo,Robert Siddaway,Martin Burkert,Stefanie Stallard,Tingting Qin,Brendan Mullan,Ruby Siada,Ramya Ravindran,Michael Niculcea,Abigail Dowling,Joshua Bradin,Kevin F. Ginn,Melissa Gener,Kathleen Dorris,Nicholas A Vitanza,Susanne Schmidt,Jasper Spitzer,Li Jiang,Mariella G. Filbin,Xuhong Cao,Maria G. Castro,Pedro R. Lowenstein,Rajen Mody,Arul M. Chinnaiyan,Pierre-Yves Desprez,Sean D. McAllister,Matthew D. Dun,Cynthia Hawkins,Sebastian M. Waszak,Sriram Venneti,Carl Koschmann,Viveka Nand Yadav +37 more
TL;DR: H3K27M-mediated re-activation of ID1 in DMG results in a SPARCL1+ migratory transcriptional program that is therapeutically targetable with CBD, finding that a clinical trial to standardize dosing may be beneficial.
Posted ContentDOI
Therapeutic reversal of prenatal pontine ID1 signaling in DIPG
Viveka Yadav,Micah Harris,Messinger D,Chase Thomas,Cummings,Tao Yang,Rinette Woo,Robert Siddaway,Burkert M,Stefanie Stallard,Tingting Qin,Brendan Mullan,Ruby Siada,Ramya Ravindran,Michael Niculcea,Kevin Ginn,Melissa Gener,Kathleen Dorris,Nicholas A Vitanza,Susanne V. Schmidt,Jasper Spitzer,Jiang Li,Mariella G. Filbin,Xuhong Cao,Maria G. Castro,Pedro R. Lowenstein,Rajen Mody,Chinnaiyan A,Pierre-Yves Desprez,Sandra S. McAllister,Cynthia Hawkins,Sebastian M. Waszak,Sriram Venneti,Carl Koschmann +33 more
TL;DR: In this article, the authors developed an in- utero electroporation (IUE) murine H3K27M-driven tumor model, which demonstrates increased ID1 expression in H3k27m-and ACVR1-mutated tumor cells.
