Journal ArticleDOI
Genome-wide association study identifies eight new susceptibility loci for atopic dermatitis in the Japanese population
Tomomitsu Hirota,Atsushi Takahashi,Michiaki Kubo,Tatsuhiko Tsunoda,Kaori Tomita,Masafumi Sakashita,Takechiyo Yamada,Shigeharu Fujieda,Shota Tanaka,Satoru Doi,Akihiko Miyatake,Tadao Enomoto,Chiharu Nishiyama,Nobuhiro Nakano,Keiko Maeda,Ko Okumura,Hideoki Ogawa,Shigaku Ikeda,Emiko Noguchi,Tohru Sakamoto,Nobuyuki Hizawa,Koji Ebe,Hidehisa Saeki,Takashi Sasaki,Tamotsu Ebihara,Masayuki Amagai,Satoshi Takeuchi,Masutaka Furue,Yusuke Nakamura,Mayumi Tamari +29 more
TL;DR: The findings advance the understanding of the genetic basis of atopic dermatitis and replicate the associations of the FLG, C11orf30, TMEM232-SLC25A46, TNFRSF6B-ZGPAT, OVOL1, ACTL9 and KIF3A-IL13 loci that were previously reported in GWAS of European and Chinese individuals.
Journal ArticleDOI
Engineering of the major house dust mite allergen Der f 2 for allergen-specific immunotherapy.
Toshiro Takai,Toyokazu Yokota,Masaaki Yasue,Chiharu Nishiyama,Toshifumi Yuuki,Akio Mori,Hirokazu Okudaira,Yasushi Okumura +7 more
TL;DR: The engineered allergen, in which the disulfide bond that linked the N- and C-terminal sequences of Der f 2 was disrupted, retained T- cell epitopes essential for immunotherapy and ability to stimulate T-cell proliferation.
Journal ArticleDOI
Taking control over control: use of product sensing in single cells to remove flux control at key enzymes in biosynthesis pathways.
Georg Schendzielorz,Martin Dippong,Alexander Grünberger,Dietrich Kohlheyer,Ayako Yoshida,Stephan Binder,Chiharu Nishiyama,Makoto Nishiyama,Michael Bott,Lothar Eggeling +9 more
TL;DR: In vivo screening of enzyme libraries by using metabolite sensors is extremely well suited to identify high-performance muteins required for overproduction, thus enabling ultrahigh-throughput screens by FACS.
Journal ArticleDOI
Solution structure of Der f 2, the major mite allergen for atopic diseases.
Saori Ichikawa,Hideki Hatanaka,Toshifumi Yuuki,Namiko Iwamoto,Soichi Kojima,Chiharu Nishiyama,Kenji Ogura,Yasushi Okumura,Fuyuhiko Inagaki +8 more
TL;DR: The tertiary structure of Der f 2 was determined by multidimensional nuclear magnetic resonance spectroscopy and was found to be a single-domain protein of immunoglobulin fold, and its structure was the most similar to those of the two regulatory domains of transglutaminase.
Journal ArticleDOI
Inhibitory Effect of Polyphenol-Enriched Apple Extracts on Mast Cell Degranulation in Vitro Targeting the Binding between IgE and FcεRI
Tomoko Tokura,Nobuhiro Nakano,Tomonobu Ito,Hironori Matsuda,Yoko Nagasako-Akazome,Tomomasa Kanda,Mitsuo Ikeda,Ko Okumura,Hideoki Ogawa,Chiharu Nishiyama +9 more
TL;DR: The results indicate that the inhibition of binding between FcεRI and IgE by either CAP or ACT was the probable cause of the suppression of mast cell activation.
