Journal ArticleDOI

Biochemical characterization of the active heterodimer form of human heparanase (Hpa1) protein expressed in insect cells.

TL;DR: An insect cell expression system used for the generation of large amounts of recombinant protein of high specific activity is presented here for the first time and it is confirmed that the 8 kDa subunit is essential for enzyme activity.
Journal ArticleDOI

Structural basis for recognition of synaptic vesicle protein 2C by botulinum neurotoxin A

TL;DR: The high-resolution crystal structure of the BoNT/A receptor-binding domain (BoNT/ a-RBD) in complex with the SV2C luminal domain (SV2C-LD) is determined, providing a strong platform for the development of novel antitoxin agents and for the rational design of Bo NT/A variants with improved therapeutic properties.
Journal ArticleDOI

Epitope determines efficacy of therapeutic anti-Tau antibodies in a functional assay with human Alzheimer Tau

TL;DR: A robust and quantitative cell based assay was developed and an unbiased screening approach was employed to identify the antibody with the highest activity against human Tau seeds, which potently blocked the seeding of human AD Tau and was fully efficacious against seeds from progressive supranuclear palsy.
Journal ArticleDOI

Small molecules that inhibit TNF signalling by stabilising an asymmetric form of the trimer.

TL;DR: This discovery paves the way for a class of small molecule drugs capable of modulating TNF function by stabilising a naturally sampled, receptor-incompetent conformation of TNF, and may prove to be a more general mechanism for inhibiting protein–protein interactions.
