Journal ArticleDOI
Enantioselective Enzymes by Computational Design and In Silico Screening
TL;DR: A strategy to design small mutant libraries of complementary enantioselective epoxide hydrolase variants for the production of highly enantioenriched (S,S)- diols and (R,R)-diols is developed.
Journal ArticleDOI
Cometabolic degradation of trichloroethylene by Pseudomonas cepacia G4 in a chemostat with toluene as the primary substrate.
Andrew S. Landa,E.M Sipkema,Jan Weijma,Antonie A. C. M. Beenackers,Jan Dolfing,Dick B. Janssen +5 more
TL;DR: Results show that P. cepacia can stably and continuously degrade toluene and TCE simultaneously in a single-reactor system without biomass retention and that the organism is more resistant to high concentrations and shock loadings of TCE than Methylosinus trichosporium OB3b.
Journal ArticleDOI
Histidine 289 Is Essential for Hydrolysis of the Alkyl-enzyme Intermediate of Haloalkane Dehalogenase
F Pries,Jaap Kingma,Geja H. Krooshof,C. Margot Jeronimus-Stratingh,Andries P. Bruins,Dick B. Janssen +5 more
TL;DR: The results show that His is the base catalyst for the dealkylation of the covalent intermediate, but that it is not essential for the initial nucleophilic attack of Asp on the C-1 atom of the haloalkane.
Journal ArticleDOI
Discovery of a Novel Styrene Monooxygenase Originating from the Metagenome
TL;DR: The newly identified monooxygenase catalyzes the epoxidation of styrene and styrene derivatives and forms the corresponding (S)-epoxides with excellent enantiomeric excess and therefore is named styrene monooxgenase subunit A (SmoA), which shows high enantioselectivity towards aromatic sulfides.
