Novel Cell- and Tissue-Based Assays for Detecting Misfolded and Aggregated Protein Accumulation Within Aggresomes and Inclusion Bodies
Dee Shen,Jack Coleman,Eric Chan,Thomas P. Nicholson,Lijun Dai,Paul W. Sheppard,Wayne F. Patton +6 more
TL;DR: A homogenous fluorescence-based assay was devised to detect aggregated proteins inside aggresomes and inclusion bodies within an authentic cellular context, allowing assessment of the effects of protein aggregation directly in cells, without resorting to the use of non-physiological protein mutations or genetically engineered cell lines.
Journal ArticleDOI
A novel image-based cytometry method for autophagy detection in living cells
Leo Li-Ying Chan,Dee Shen,Alisha R. Wilkinson,Wayne F. Patton,Ning Lai,Eric Chan,Dmitry Kuksin,Bo Lin,Jean Qiu +8 more
TL;DR: A novel method using the Cellometer image-based cytometry/fluorescent dye method in combination with Cyto-ID® Green dye for autophagy detection in live cells is developed and compared with flow cytometry by measuring macroautophagy in nutrient-starved Jurkat cells.
Journal ArticleDOI
Role of a Contactin multi‐molecular complex secreted by oligodendrocytes in nodal protein clustering in the CNS
Anne-Laure Dubessy,Elisa Mazuir,Quentin Rappeneau,Sokounthie Ou,Charly Abi Ghanem,Kevin Piquand,Marie-Stéphane Aigrot,Melina Thetiot,Anne Desmazières,Eric Chan,Matt Fitzgibbon,Mark D. Fleming,Raul Krauss,Bernard Zalc,Barbara Ranscht,Catherine Lubetzki,Nathalie Sol-Foulon +16 more
TL;DR: The results identify Contactin‐1 secreted by oligodendrocytes as a novel factor that may influence early steps of nodal sodium channel cluster formation along specific axon populations.
Proceedings ArticleDOI
Abstract 5346: A novel immunotoxin comprising quadruple RNase tethered to an internalizing anti-TROP-2 humanized MAb shows potent cytotoxicity against diverse solid tumorsin vitro
Thomas M. Cardillo,Edmund A. Rossi,Roberto Arrojo,Eric Chan,David M. Goldenberg,Chien-Hsing Chang +5 more
TL;DR: The cell binding data obtained for these solid cancer cell lines suggest that the sensitivity of a cell line to E1-Rap appears to correlate with its TROP-2 expression on the cell surface, which is 1,000- to 100,00-fold higher than untargeted Rap or the combination of Rap and hRS7.
Journal ArticleDOI
CRISPR/Cas9 Genome Editing to Treat Sickle Cell Disease and B-Thalassemia: Re-Creating Genetic Variants to Upregulate Fetal Hemoglobin Appear Well-Tolerated, Effective and Durable.
Michelle I. Lin,Elizabeth J. Paik,Bibhu P. Mishra,David Burkhardt,Andrew Kernytsky,Michael A. Pettiglio,Yi-Shan Chen,Kaleigh Tomkinson,Amanda Woo,Mauricio Cortes,Siyuan Tan,Todd D Borland,Lawrence Klein,Angela Yen,Sudipta Mahajan,Eric Chan,Brenda K. Eustace,Matthew H. Porteus,Tirtha Chakraborty,Chad A. Cowan,Rodger Novak,Ante Sven Lundberg +21 more
TL;DR: Findings show that a CRISPR/Cas9-based therapy has the potential to upregulate HbF and treat diseases of b-globin and the long-term persistence of transplanted cells in a NOD-SCID-Gamma xeno-transplant mouse model is demonstrated.
