A Nine-transmembrane Domain Topology for Presenilin 1
Hanna Laudon,Emil M. Hansson,Karin Melén,Anna Bergman,Mark R. Farmery,Bengt Winblad,Urban Lendahl,Gunnar von Heijne,Jan Näslund +8 more
TL;DR: Glycosylation acceptor sequences were introduced into full-length human PS1, and the results showed that the first hydrophilic loop is oriented toward the lumen of the endoplasmic reticulum, whereas the N terminus and large hydrophobic loop are in the cytosol.
Journal ArticleDOI
The murine version of BAN2401 (mAb158) selectively reduces amyloid-β protofibrils in brain and cerebrospinal fluid of tg-ArcSwe mice.
Stina Tucker,Christer Möller,Karin Tegerstedt,Anna Lord,Hanna Laudon,Johan Sjödahl,Linda Söderberg,Erika Spens,Charlotte Sahlin,Erik Rollman Waara,Andrew Satlin,Pär Gellerfors,Gunilla Osswald,Lars Lannfelt +13 more
TL;DR: It is demonstrated that the humanized antibody BAN2401 and its murine version mAb158 exhibit a strong binding preference for Aβ protofibrils over Aβ monomers, and further support is provided in favor of its position as an attractive new candidate for AD immunotherapy.
Journal ArticleDOI
Presenilin-1 Mutation L271V Results in Altered Exon 8 Splicing and Alzheimer's Disease with Non-cored Plaques and No Neuritic Dystrophy
John B.J. Kwok,Glenda M. Halliday,Glenda M. Halliday,William S. Brooks,William S. Brooks,Georgia Dolios,Hanna Laudon,Ohoshi Murayama,Marianne Hallupp,Renee F. Badenhop,Renee F. Badenhop,James C. Vickers,Rong Wang,Jan Näslund,Akihiko Takashima,Sam Gandy,Peter R. Schofield,Peter R. Schofield +17 more
TL;DR: It is postulate that variant plaques observed in this family are due in part to the effects of PS-1Δexon8 and that interaction between PS- 1 and various protein complexes are necessary for neuritic plaque formation.
Journal ArticleDOI
Co-expressed presenilin 1 NTF and CTF form functional γ-secretase complexes in cells devoid of full-length protein
Hanna Laudon,Paul M. Mathews,Helena Karlström,Anna Bergman,Mark R. Farmery,Ralph A. Nixon,Bengt Winblad,Sam Gandy,Urban Lendahl,Johan Lundkvist,Jan Näslund +10 more
TL;DR: It is demonstrated that coexpression of PS1 NTF and CTF functionally mimics expression of the full‐length PS1 protein and restores γ‐secretase activity in PS‐deficient mammalian cells.
Journal ArticleDOI
The Extreme C Terminus of Presenilin 1 Is Essential for γ-Secretase Complex Assembly and Activity
TL;DR: The effects of modifications on the C terminus of PS1 on PS1 endoproteolysis, γ-secretase complex assembly, and activity in cells devoid of endogenous PS are investigated and suggest that the PS1 N- and C-terminal fragment intermolecular interactions are independent of an association with nicastrin and Aph-1.
