Cholesterol-functionalized DNA/RNA heteroduplexes cross the blood–brain barrier and knock down genes in the rodent CNS
Tetsuya Nagata,Chrissa A. Dwyer,Kie Yoshida-Tanaka,Kensuke Ihara,Masaki Ohyagi,Hidetoshi Kaburagi,Haruka Miyata,Satoe Ebihara,Kotaro Yoshioka,Takashi Ishii,Kanjiro Miyata,Kenichi Miyata,Berit Powers,Tomoko Igari,Syunsuke Yamamoto,Naoto Arimura,Hideki Hirabayashi,Toshiki Uchihara,Rintaro Iwata Hara,Rintaro Iwata Hara,Takeshi Wada,C. Frank Bennett,Punit P. Seth,Frank Rigo,Takanori Yokota +24 more
TL;DR: In this article, DNA/RNA heteroduplex oligonucleotides conjugated to cholesterol or α-tocopherol at the 5′ end of the RNA strand reach the central nervous system (CNS) after subcutaneous or intravenous administration in mice and rats.
Journal ArticleDOI
Intrathecal AAV Serotype 9-mediated Delivery of shRNA Against TRPV1 Attenuates Thermal Hyperalgesia in a Mouse Model of Peripheral Nerve Injury
Takashi Hirai,Mitsuhiro Enomoto,Hidetoshi Kaburagi,Shinichi Sotome,Kie Yoshida-Tanaka,Madoka Ukegawa,Hiroya Kuwahara,Mariko Yamamoto,Mio Tajiri,Haruka Miyata,Yukihiko Hirai,Makoto Tominaga,Kenichi Shinomiya,Hidehiro Mizusawa,Atsushi Okawa,Takanori Yokota +15 more
TL;DR: This study provides important evidence for the contribution of TRPV1 to thermal hypersensitivity in neuropathic pain and establishes intrathecal AAV9-mediated gene delivery as an investigative and potentially therapeutic platform for the nervous system.
Journal ArticleDOI
Angubindin-1 opens the blood-brain barrier in vivo for delivery of antisense oligonucleotide to the central nervous system.
Satoshi Zeniya,Hiroya Kuwahara,Kaiichi Daizo,Akihiro Watari,Masuo Kondoh,Kie Yoshida-Tanaka,Hidetoshi Kaburagi,Ken Asada,Tetsuya Nagata,Masahiro Nagahama,Kiyohito Yagi,Takanori Yokota +11 more
TL;DR: This proof‐of‐concept study demonstrated that intravenously injected angubindin‐1 increased the permeability of the blood–brain barrier and enabled transient delivery of subsequently administered antisense oligonucleotides into the mouse brain and spinal cord, leading to silencing of a target RNA without any overt adverse effects.
Journal ArticleDOI
Bone Marrow Stromal Cells Combined With a Honeycomb Collagen Sponge Facilitate Neurite Elongation In Vitro and Neural Restoration in the Hemisected Rat Spinal Cord.
Madoka Onuma-Ukegawa,Kush Bhatt,Takashi Hirai,Hidetoshi Kaburagi,Shinichi Sotome,Yoshiaki Wakabayashi,Shizuko Ichinose,Kenichi Shinomiya,Atsushi Okawa,Mitsuhiro Enomoto +9 more
TL;DR: The study indicates that the application of BMSCs in a HC scaffold in the injured spinal cord directly promoted sensory nerve and rubrospinal tract regeneration, thus resulting in functional recovery.
Journal ArticleDOI
Lower grip strength and dynamic body balance in women with distal radial fractures
Koji Fujita,Hidetoshi Kaburagi,Akimoto Nimura,Takashi Miyamoto,Yoshiaki Wakabayashi,Y. Seki,H. Aoyama,Haruhiko Shimura,R. Kato,Atsushi Okawa +9 more
TL;DR: It was concluded that women with distal radial fractures who were surgically treated showed lower grip strength and dynamic body balancing than those of controls, suggesting that measurements of grip strengthand dynamic body balance may be useful screening tools to assess future fracture risk.
