Journal ArticleDOI
Integrative Epigenetic and Gene Expression Analysis of Renal Tumor Progression to Metastasis
Hyeyoung Nam,Darshan S. Chandrashekar,Anirban Kundu,Sandeep B. Shelar,Eun-Young Kho,Guru Sonpavde,Gurudatta Naik,Pooja Ghatalia,Carolina B. Livi,Sooryanarayana Varambally,Sooryanarayana Varambally,Sunil Sudarshan,Sunil Sudarshan +12 more
TL;DR: Collectively, these data provide significant insight into the biology of aggressive RCC and demonstrate a novel role for DNA methylation in the promotion of HIF signaling and invasive phenotypes in renal cancer.
Journal ArticleDOI
PRDM16 suppresses HIF-targeted gene expression in kidney cancer.
Anirban Kundu,Hyeyoung Nam,Sandeep B. Shelar,Darshan S. Chandrashekar,Garrett J. Brinkley,Suman Karki,Tanecia Mitchell,Carolina B. Livi,Phillip Buckhaults,Richard Kirkman,Yawen Tang,Glenn C. Rowe,Shi Wei,Sooryanarayana Varambally,Sunil Sudarshan,Sunil Sudarshan +15 more
TL;DR: The transcription factor PRDM16 is epigenetically silenced in kidney cancer and loss of this factor’s repressive properties promotes HIF-mediated induction of the protumorigenic factor SEMA5B, which provides a novel epigenetic mechanism by which HIF signaling is promoted in this malignancy.
Journal ArticleDOI
PGC1α suppresses kidney cancer progression by inhibiting collagen-induced SNAIL expression.
Hyeyoung Nam,Anirban Kundu,Garrett J. Brinkley,Darshan S. Chandrashekar,Richard Kirkman,Balabhadrapatruni V. S. K. Chakravarthi,Rachael M. Orlandella,Lyse A. Norian,Guru Sonpavde,Pooja Ghatalia,Fei Fei,Shi Wei,Sooryanarayana Varambally,Sunil Sudarshan,Sunil Sudarshan +14 more
TL;DR: A novel role for PGC1α in the regulation of proinvasive SNAIL proteins is demonstrated and this work reports that the decreased expression of peroxisome proliferator-activated receptor gamma, coactivator 1 alpha and the increased expression of several genes encoding collagen family members are associated with RCC tumor progression.
Journal ArticleDOI
14-3-3 proteins protect AMPK-phosphorylated ten-eleven translocation-2 (TET2) from PP2A-mediated dephosphorylation
Anirban Kundu,Sandeep B. Shelar,Arindam P. Ghosh,Mary E. Ballestas,Richard Kirkman,Hyeyoung Nam,Garrett J. Brinkley,Suman Karki,James A. Mobley,Sejong Bae,Sooryanarayana Varambally,Sunil Sudarshan,Sunil Sudarshan +12 more
TL;DR: It is observed that AMP-activated protein kinase–mediated phosphorylation at Ser-99 promotes TET2 stability and increases global DNA 5-hydroxymethylcytosine levels and that protein phosphatase 2A can interact with Tet2 and dephosphorylate Ser- 99.
Journal ArticleDOI
Teleological role of L-2-hydroxyglutarate dehydrogenase in the kidney
Garrett J. Brinkley,Hyeyoung Nam,Eun-Hee Shim,Richard Kirkman,Anirban Kundu,Suman Karki,Yasaman Heidarian,Jason M. Tennessen,Juan Liu,Jason W. Locasale,Tao Guo,Shi Wei,Jennifer B. Gordetsky,Teresa L. Johnson-Pais,Devin Absher,Dinesh Rakheja,Anil K. Challa,Sunil Sudarshan,Sunil Sudarshan +18 more
TL;DR: A novel role for L-2-hydroxyglutarate dehydrogenase in the kidney is demonstrated as well as new insight into the mechanisms that regulate expression of this enzyme.
