Abstract: Thyroid-stimulating hormone increased adenyl cyclase activity in canine and bovine thyroid homogenates. Activation of ade-nyl cyclase was impaired by 3 adrenergic blocking agents, dl-propranolol, d-propranolol and phentolamine, over the range 3−7.5 × 10−4m. The concentrations are approximately 100 times greater than those which produce alpha- or beta-adrenergic blockade. dl-Propranolol and phen-tolamine at 1.5−3.0 × 10−4M impaired thyroid-stimulating hormone stimulation of glucose-l-14C oxidation in canine thyroid slices. A similar effect was seen when the slices were preincubated with propranolol, washed, and then incubated with thyroid-stimulating hormone in the absence of propranolol. Propranolol and phentolamine at concentrations which abolished thyroid-stimulating hormone stimulated glucose oxidation in canine thyroid slices had little effect on the stimulation of glucose oxidation by dibutyryl cyclic 3′,5′-AMP.
