Journal ArticleDOI
Selective Sirt2 inhibition by ligand-induced rearrangement of the active site.
Tobias Rumpf,Matthias Schiedel,Berin Karaman,Claudia Roessler,Brian J. North,Attila Lehotzky,Judit Oláh,Kathrin I. Ladwein,Karin Schmidtkunz,Markus Gajer,Martin Pannek,Clemens Steegborn,David A. Sinclair,Stefan Gerhardt,Judit Ovádi,Mike Schutkowski,Wolfgang Sippl,Oliver Einsle,Manfred Jung +18 more
TL;DR: High-resolution structures of human Sirt2 in complex with highly selective drug-like inhibitors that show a unique inhibitory mechanism are presented and structural insights into this unique mechanism of selective sirtuin inhibition provide the basis for further inhibitor development and selective tools for sIRTuin biology.
Journal ArticleDOI
Chemically Induced Degradation of Sirtuin 2 (Sirt2) by a Proteolysis Targeting Chimera (PROTAC) Based on Sirtuin Rearranging Ligands (SirReals)
Matthias Schiedel,Daniel Herp,Sören Hammelmann,Sören Swyter,Attila Lehotzky,Dina Robaa,Judit Oláh,Judit Ovádi,Wolfgang Sippl,Manfred Jung +9 more
TL;DR: This SirReal-based PROTAC is the first example of a probe that is able to chemically induce the degradation of an epigenetic eraser protein and can be readily adapted to alkynylated ligands of other targets.
Journal ArticleDOI
Clinical performance of the Nevisense system in cutaneous melanoma detection: an international, multicentre, prospective and blinded clinical trial on efficacy and safety.
Joseph Malvehy,Axel Hauschild,Clara Curiel-Lewandrowski,Peter Mohr,Rainer Hofmann-Wellenhof,Richard Motley,Carola Berking,Douglas Grossman,John Paoli,Carmen Loquai,Judit Oláh,Uwe Reinhold,H. Wenger,Thomas Dirschka,S. Davis,Catriona Henderson,H. Rabinovitz,Julia Welzel,Dirk Schadendorf,Ulrik Birgersson +19 more
TL;DR: The Nevisense electrical impedance spectroscopy (EIS) system (SciBase AB) was designed and shown to have the potential to be used as an adjunct diagnostic tool for melanoma detection.
Journal ArticleDOI
Triosephosphate isomerase deficiency: new insights into an enigmatic disease
TL;DR: This paper reviews the recent studies on the structural and catalytic changes caused by mutation and/or nitrotyrosination of the isomerase leading to the formation of an aggregation-prone protein, a characteristic of conformational disorders.
Journal ArticleDOI
Interactions of Pathological Hallmark Proteins: TUBULIN POLYMERIZATION PROMOTING PROTEIN/p25, β-AMYLOID, AND α-SYNUCLEIN*
Judit Oláh,Orsolya Vincze,Dezső P. Virok,Dóra Simon,Zsolt Bozsó,Natália Tőkési,István T. Horváth,Emma Hlavanda,János Kovács,Anna Magyar,Mária Szűcs,Mária Szűcs,Ferenc Orosz,Botond Penke,Botond Penke,Judit Ovádi +15 more
TL;DR: The finding that the interaction of TPPP/p25 with Aβ can produce pathological-like aggregates is tightly coupled with unusual pathology of the Alzheimer disease revealed previously; that is, partial co-localization of Aβ and TPPP /p25 in the case of diffuse Lewy body disease with Alzheimer disease.
