Privileged substructures are of potentially great importance in medicinal chemistry. These scaffolds are characterized by their ability to promiscuously bind to a multitude of receptors through a variety of favorable characteristics. This may include presentation of their substituents in a spatially defined manner and perhaps also the ability to directly bind to the receptor itself, as well as exhibiting promising characteristics to aid bioavailability of the overall molecule. It is believed that some privileged substructures achieve this through the mimicry of common protein surface elements that are responsible for binding, such as β- and gamma;-turns. As a result, these structures represent a promising means by which new lead compounds may be identified.

Recent Developments in Ring Opening Polymerization of Lactones for Biomedical Applications

Polycaprolactone (PCL) is an important polymer due to its mechanical properties, miscibility with a large range of other polymers and biodegradability. Two main pathways to produce polycaprolactone have been described in the literature: the polycondensation of a hydroxycarboxylic acid: 6-hydroxyhexanoic acid, and the ring-opening polymerisation (ROP) of a lactone: e-caprolactone (e-CL). This critical review summarises the different conditions which have been described to synthesise PCL, and gives a broad overview of the different catalytic systems that were used (enzymatic, organic and metal catalyst systems). A surprising variety of catalytic systems have been studied, touching on virtually every section of the periodic table. A detailed list of reaction conditions and catalysts/initiators is given and reaction mechanisms are presented where known. Emphasis is put on the ROP pathway due to its prevalence in the literature and the superior polymer that is obtained. In addition, ineffective systems that have been tried to catalyse the production of PCL are included in the electronic supplementary information for completeness (141 references).

Biodegradable synthetic polymers: Preparation, functionalization and biomedical application

The fatty acid amides are a family of lipids composed of two chemical moieties, a fatty acid and a biogenic amine linked together in an amide bond. This lipid family is structurally related to the endocannabinoid anandamide (N-arachidonoylethanolamine) and, thus, is frequently referred to as a family of endocannabinoid-related lipids. The fatty acid amide family is divided into different classes based on the conjugate amine; anandamide being a member of the N-acylethanolamine class (NAE). Another class within the fatty acid amide family is the N-acyl amino acids (NA-AAs). The focus of this review is a sub-class of the NA-AAs, the N-acyl aromatic amino acids (NA-ArAAs). The NA-ArAAs are not broadly recognized, even by those interested in the endocannabinoids and endocannabinoid-related lipids. Herein, the NA-ArAAs that have been identified from a biological source will be highlighted and pathways for their biosynthesis, degradation, enzymatic modification, and transport will be presented. Also, information about the cellular functions of the NA-ArAAs will be placed in context with the data regarding the identification and metabolism of these N-acylated amino acids. A review of the current state-of-knowledge about the NA-ArAAs is to stimulate future research about this underappreciated sub-class of the fatty acid amide family.

The Biosynthesis and Metabolism of the N-Acylated Aromatic Amino Acids: N-Acylphenylalanine, N-Acyltyrosine, N-Acyltryptophan, and N-Acylhistidine

The present invention concerns farnesylamine derivatives, compositions containing these compounds, and methods of using these compounds and compositions as inhibitors of ras-mediated signal transduction and inhibitors of aberrant cell growth, e.g., as anti-cancer agents, as well as anti-fungal agents. Other non-malignant diseases characterized by proliferation of cells that may be treated using the farneylamine derivatives of the include, but are not limited to, cirrhosis of the liver; graft rejection; restenosis; and disorders characterized by a proliferation of T cells such as autoimmune diseases, e.g., type 1 diabetes, lupus and multiple sclerosis.

Mossbauer investigation on the kinetics of the pyrolysis of iron(iii) tetraphenylporphyrin chloride

The yeast lipase Candida cylindracea (CCL) and porcine pancreatic lipase (PPL) have been used for regioselective deacylation of peracetylated benzopyrones, diphenylpropenones and acetophenones for the first time. The deacylation study on different classes of polyphenols has revealed that the presence of carbonyl group attached to the aromatic ring is needed by the lipases to exhibit regioselectivity towards hydrolysis of acetoxyl groups. The acetoxyl groups at positions other than the one at ortho position to the carbonyl group get selectively hydrolysed by PPL in orgnnic solvents. The trnnsesterification reactions using triflouroethylbutyrate (TFEB), catalysed by PPL and CCL on some polyols in dry organic solvents were also performed. It was found that the primary hydroxyl is acylated. In D-panthenol, the oxidised dextrorotatory form of which is a major constituent of vitamin B-complex, the primary hydroxyl group at the far end of the asymmetric carbon atom gets exclusively acylated. This work should be of importance in the synthesis of building blocks of biologically active natural products which may provide structural leads to anti AIDS and anticancer agents.
