Journal ArticleDOI
Autophagy is important in islet homeostasis and compensatory increase of beta cell mass in response to high-fat diet
Chie Ebato,Toyoyoshi Uchida,Masayuki Arakawa,Masaaki Komatsu,Masaaki Komatsu,Masaaki Komatsu,Takashi Ueno,Koji Komiya,Kosuke Azuma,Takahisa Hirose,Keiji Tanaka,Eiki Kominami,Ryuzo Kawamori,Yoshio Fujitani,Hirotaka Watada +14 more
TL;DR: Upregulation of autophagosome formation in pancreatic beta cells in diabetic db/db and in nondiabetic high-fat-fed C57BL/6 mice suggests that basal autophagy is important for maintenance of normal islet architecture and function.
Journal ArticleDOI
Gut Dysbiosis and Detection of “Live Gut Bacteria” in Blood of Japanese Patients With Type 2 Diabetes
Junko Sato,Akio Kanazawa,Fuki Ikeda,Tomoaki Yoshihara,Hiromasa Goto,Hiroko Abe,Koji Komiya,Minako Kawaguchi,Tomoaki Shimizu,Takeshi Ogihara,Yoshifumi Tamura,Yuko Sakurai,Risako Yamamoto,Tomoya Mita,Yoshio Fujitani,Hiroshi Fukuda,Koji Nomoto,Takuya Takahashi,Takashi Asahara,Takahisa Hirose,Satoru Nagata,Yuichiro Yamashiro,Hirotaka Watada +22 more
TL;DR: This is the first report of gut dysbiosis in Japanese patients with type 2 diabetes as assessed by RT-qPCR, and the high rate of gut bacteria in the circulation suggests translocation of bacteria from the gut to the bloodstream.
Journal ArticleDOI
Human IAPP-induced pancreatic β cell toxicity and its regulation by autophagy.
Nayumi Shigihara,Ayako Fukunaka,Akemi Hara,Koji Komiya,Akira Honda,Toyoyoshi Uchida,Hiroko Abe,Yukiko Toyofuku,Motoyuki Tamaki,Takeshi Ogihara,Takeshi Miyatsuka,Henry J. Hiddinga,Setsuya Sakagashira,Masato Koike,Yasuo Uchiyama,Tamotsu Yoshimori,Tamotsu Yoshimori,Norman L. Eberhardt,Yoshio Fujitani,Hirotaka Watada +19 more
TL;DR: Treatment of INS-1 cells with human IAPP enhances cell death, inhibits cytoproliferation, and increases autophagosome formation, and inhibition of autophagy increased the vulnerability of β cells to the cytotoxic effects of hIAPP.
Journal ArticleDOI
Free fatty acids stimulate autophagy in pancreatic β-cells via JNK pathway.
Koji Komiya,Toyoyoshi Uchida,Takashi Ueno,Masato Koike,Hiroko Abe,Takahisa Hirose,Ryuzo Kawamori,Yasuo Uchiyama,Eiki Kominami,Yoshio Fujitani,Hirotaka Watada +10 more
TL;DR: The results confirmed that palmitate activates autophagic flux in most of the cells, and suggested that FFA stimulated functional autophagy possibly through the PKR-JNK1 pathway independent of ER or oxidative stress.
Journal ArticleDOI
Exendin-4, a glucagon-like peptide-1 receptor agonist, reduces intimal thickening after vascular injury
Hiromasa Goto,Takashi Nomiyama,Tomoya Mita,Eisuke Yasunari,Kosuke Azuma,Koji Komiya,Masayuki Arakawa,Wen Long Jin,Akio Kanazawa,Ryuzo Kawamori,Yoshio Fujitani,Takahisa Hirose,Hirotaka Watada +12 more
TL;DR: It is suggested that exendin-4 reduced intimal thickening after vascular injury at least in part by the suppression of platelet-derived growth factor-induced smooth muscle cells proliferation.
