Journal ArticleDOI
Loss-of-function mutations in Notch receptors in cutaneous and lung squamous cell carcinoma.
Nicholas J. Wang,Zachary Sanborn,Kelly L. Arnett,Laura J. Bayston,Wilson Liao,Charlotte M. Proby,Irene M. Leigh,Eric A. Collisson,Patricia B. Gordon,Lakshmi Jakkula,Sally D. Pennypacker,Yong Zou,Mimansa Sharma,Jeffrey P. North,Swapna S. Vemula,Theodora M. Mauro,Isaac M. Neuhaus,Philip E. LeBoit,Joe S Hur,Kyunghee Park,Nam Huh,Pui-Yan Kwok,Sarah T. Arron,Pierre P. Massion,Allen E. Bale,David Haussler,James E. Cleaver,Joe W. Gray,Paul T. Spellman,Andrew P. South,Jon C. Aster,Stephen C. Blacklow,Stephen C. Blacklow,Raymond J. Cho +33 more
TL;DR: The bifunctional role of Notch in human cancer thus emphasizes the context dependency of signaling outcomes and suggests that targeted inhibition of the Notch pathway may induce squamous epithelial malignancies.
Journal ArticleDOI
Temporal Dissection of Tumorigenesis in Primary Cancers
Steffen Durinck,Christine Ho,Nicholas J. Wang,Wilson Liao,Lakshmi Jakkula,Eric A. Collisson,Jennifer Pons,Sai Wing Chan,Ernest T. Lam,Catherine Chu,Kyunghee Park,Sung-Woo Hong,Joe S Hur,Nam Huh,Isaac M. Neuhaus,Siegrid S. Yu,Roy C. Grekin,Theodora M. Mauro,James E. Cleaver,Pui-Yan Kwok,Philip E. LeBoit,Gad Getz,Kristian Cibulskis,Jon C. Aster,Haiyan Huang,Elizabeth Purdom,Jian Li,Jian Li,Lars Bolund,Lars Bolund,Sarah T. Arron,Joe W. Gray,Paul T. Spellman,Raymond J. Cho +33 more
TL;DR: This work integrates DNA sequence and copy number information to reconstruct the order of abnormalities as individual tumors evolve for 2 separate cancer types, and reframe paradigms in which TP53 mutation is required later, to bypass senescence induced by driver oncogenes.
Journal ArticleDOI
Prediction of recurrence-free survival in postoperative non-small cell lung cancer patients by using an integrated model of clinical information and gene expression.
Eung-Sirk Lee,Dae-Soon Son,Sung-Hyun Kim,Jinseon Lee,Jisuk Jo,Joungho Han,Heesue Kim,Hyunjoo Lee,Hye Young Choi,Youngja Jung,Miyeon Park,Yu Sung Lim,Kwhanmien Kim,Young Mog Shim,Byung-Chul Kim,Kyu-Sang Lee,Nam Huh,Christopher Ko,Kyunghee Park,Jae Won Lee,Yong Soo Choi,Yong Soo Choi,Jhingook Kim,Jhingook Kim +23 more
TL;DR: An accurate, technically simple, and reproducible method for predicting individual recurrence risks is developed and would potentially be useful in developing customized strategies for managing lung cancer.
Journal ArticleDOI
Prevalence and detection of low-allele-fraction variants in clinical cancer samples
Hyun-Tae Shin,Hyun-Tae Shin,Yoon-La Choi,Yoon-La Choi,Jae Won Yun,Jae Won Yun,Nayoung K.D. Kim,Sook-Young Kim,Hyo Jeong Jeon,Jae-Yong Nam,Jae-Yong Nam,Chung Lee,Chung Lee,Daeun Ryu,Daeun Ryu,Sang Cheol Kim,Kyunghee Park,Eunjin Lee,Joon Seol Bae,Dae-Soon Son,Je-Gun Joung,Jeeyun Lee,Seung Tae Kim,Myung-Ju Ahn,Se-Hoon Lee,Jin Seok Ahn,Woo Yong Lee,Bo Young Oh,Bo Young Oh,Yeon Hee Park,Jeong Eon Lee,Kwang Hyuk Lee,Hee Cheol Kim,Kyoung-Mee Kim,Young-Hyuck Im,Keunchil Park,Peter J. Park,Woong-Yang Park,Woong-Yang Park +38 more
TL;DR: It is demonstrated that many clinically-actionable variants have low variant allele fractions, requiring assays with high detection sensitivity, and that capturing low VAF mutations at hotspots by sufficient sequencing coverage and carefully tuned algorithms is imperative for a clinical assay.
Journal ArticleDOI
Mutational profiling of brain metastasis from breast cancer: matched pair analysis of targeted sequencing between brain metastasis and primary breast cancer
Ji Yun Lee,Kyunghee Park,Sung Hee Lim,Hae Su Kim,Kwai Han Yoo,Ki Sun Jung,Haa Na Song,Mineui Hong,In Gu Do,Tae-jin Ahn,Se Kyung Lee,Soo Youn Bae,Seok Won Kim,Jeong Eon Lee,Seok Jin Nam,Duk-Hwan Kim,Hae Hyun Jung,Ji-Yeon Kim,Jin Seok Ahn,Young-Hyuck Im,Yeon Hee Park +20 more
TL;DR: Interestingly, gene expression in BCBM such as TP53, PIK3CA, KIT, MLH1, and RB1 also seemed to be present in primary breast cancers, suggesting the biology of metastasis and its susceptibility to treatment should be understood.
