Journal ArticleDOI
5' CpG island methylation is associated with transcriptional silencing of the tumour suppressor p16/CDKN2/MTS1 in human cancers.
Adrian Merlo,James G. Herman,Li Mao,Daniel J. Lee,Edward Gabrielson,Peter C. Burger,Stephen B. Baylin,David Sidransky +7 more
TL;DR: De novo methylation of the 5′ CpG island of p16 was found in approximately 20% of different primary neoplasms, but not in normal tissues, potentially representing a common pathway of tumour suppressor gene inactivation in human cancers.
Journal Article
Inactivation of the CDKN2/p16/MTS1 Gene Is Frequently Associated with Aberrant DNA Methylation in All Common Human Cancers
James G. Herman,Adrian Merlo,Li Mao,Rena G. Lapidus,Jean Pierre J. Issa,Nancy E. Davidson,David Sidransky,Stephen B. Baylin +7 more
TL;DR: In tumors, de novo methylation of the 5' CpG island is a frequent mode of inactivation of CDKN2/p16 and this alteration of p16 in colon cancer was particularly striking, since inactivation does not occur through homozygous deletion in this tumor type.
Journal ArticleDOI
The BATTLE Trial: Personalizing Therapy for Lung Cancer
Edward S. Kim,Roy S. Herbst,Ignacio I. Wistuba,J. Jack Lee,George R. Blumenschein,Anne Tsao,David J. Stewart,Marshall E. Hicks,Jeremy J. Erasmus,Sanjay Gupta,Christine M. Alden,Suyu Liu,Ximing Tang,Fadlo R. Khuri,Hai T. Tran,Bruce E. Johnson,John V. Heymach,Li Mao,Frank V. Fossella,Merrill S. Kies,Vassiliki A. Papadimitrakopoulou,Suzanne E. Davis,Scott M. Lippman,Waun Ki Hong +23 more
TL;DR: The BATTLE study is the first completed prospective, adaptively randomized study in heavily pretreated NSCLC patients that mandated tumor profiling with "real-time" biopsies, taking a substantial step toward realizing personalized lung cancer therapy by integrating real-time molecular laboratory findings.
Journal ArticleDOI
Molecular Assessment of Histopathological Staging in Squamous-Cell Carcinoma of the Head and Neck
Joseph Brennan,Li Mao,Ralph H. Hruban,Jay O. Boyle,Yolanda Eby,Wayne M. Koch,Steven N. Goodman,David Sidransky +7 more
TL;DR: Molecular analysis of surgical margins and lymph nodes can augment standard histopathological assessment and may improve the prediction of local tumor recurrence.
Journal ArticleDOI
Frequency of homozygous deletion at p16/CDKN2 in primary human tumours
Paul Cairns,Thomas J. Polascik,Yolanda Eby,Kaori Tokino,Joseph A. Califano,Adrian Merlo,Li Mao,J. Herath,Robert B. Jenkins,William H. Westra,Joni L. Rutter,Alan Buckler,Edward Gabrielson,Melvyn S. Tockman,Kathleen R. Cho,Lora Hedrick,G S Bova,William B. Isaacs,Wayne M. Koch,Donna Schwab,David Sidransky +20 more
TL;DR: It is found that small homozygous deletions represent the predominant mechanism of inactivation at 9p21 in bladder tumours and are present in other tumour types, including breast and prostate cancer.
