TOPICAL REVIEW: Applications of magnetic nanoparticles in biomedicine

The physical principles underlying some current biomedical applications of magnetic nanoparticles are reviewed. Starting from well-known basic concepts, and drawing on examples from biology and biomedicine, the relevant physics of magnetic materials and their responses to applied magnetic fields are surveyed. The way these properties are controlled and used is illustrated with reference to (i) magnetic separation of labelled cells and other biological entities; (ii) therapeutic drug, gene and radionuclide delivery; (iii) radio frequency methods for the catabolism of tumours via hyperthermia; and (iv) contrast enhancement agents for magnetic resonance imaging applications. Future prospects are also discussed.

Superparamagnetic iron oxide nanoparticles (SPIONs): Development, surface modification and applications in chemotherapy

Proteins, through their unique and specific interactions with other macromolecules and inorganics, control structures and functions of all biological hard and soft tissues in organisms. Molecular biomimetics is an emerging field in which hybrid technologies are developed by using the tools of molecular biology and nanotechnology. Taking lessons from biology, polypeptides can now be genetically engineered to specifically bind to selected inorganic compounds for applications in nano- and biotechnology. This review discusses combinatorial biological protocols, that is, bacterial cell surface and phage-display technologies, in the selection of short sequences that have affinity to (noble) metals, semiconducting oxides and other technological compounds. These genetically engineered proteins for inorganics (GEPIs) can be used in the assembly of functional nanostructures. Based on the three fundamental principles of molecular recognition, self-assembly and DNA manipulation, we highlight successful uses of GEPI in nanotechnology.

Preparation and Characterization of Magnetic Nanoparticles

Magnetic fluids used in biomedicine have to be biocompatible and therefore the magnetic nanoparticles are modified by different biocompatible materials. In this work the magnetic nanoparticles Fe3O4 sterically stabilized by sodium oleate were prepared by coprecipitation method. Consequently they were modified with polyethylene glycol (PEG) of different molecular weights and different PEG to magnetite Fe3O4 feed weight ratios varying from 0.01 to 30 to produce biocompatible magnetic fluids (MFPEG). The morphology was observed by scanning electron microscopy. The magnetic nanoparticles coated with PEG showed almost spherical shape for all studied systems of MFPEG. Differential scanning calorimetry (DSC) was used to study the adsorption of PEG on magnetic nanoparticles and to determine the maximal amount of PEG adsorbed on the magnetic nanoparticles. The increasing PEG molecular weight leads to the decrease in maximal PEG/Fe3O4 feed weight ratio. In vitro toxicity of the magnetic fluids using cells of skin cancer of mice B16 was tested with the aim to confirm the biocompatibility of the prepared magnetic fluids.
