Repurposing of Clinically Developed Drugs for Treatment of Middle East Respiratory Syndrome Coronavirus Infection
Julie Dyall,Christopher M. Coleman,Brit J. Hart,Thiagarajan Venkataraman,Michael R. Holbrook,Jason Kindrachuk,Reed F. Johnson,Gene G. Olinger,Peter B. Jahrling,Monique Laidlaw,Lisa M. Johansen,Calli M. Lear-Rooney,Pamela J. Glass,Lisa E. Hensley,Matthew B. Frieman +14 more
TL;DR: The drugs identified in these screens provide new targets for in vivo studies as well as incorporation into ongoing clinical studies, and repurposing of existing drugs is likely the only solution for outbreaks due to emerging viruses.
Journal ArticleDOI
Antiviral Potential of ERK/MAPK and PI3K/AKT/mTOR Signaling Modulation for Middle East Respiratory Syndrome Coronavirus Infection as Identified by Temporal Kinome Analysis
Jason Kindrachuk,Britini L. Ork,Brit J. Hart,Steven Mazur,Michael R. Holbrook,Matthew B. Frieman,Dawn Traynor,Reed F. Johnson,Julie Dyall,Jens H. Kuhn,Gene G. Olinger,Lisa E. Hensley,Peter B. Jahrling +12 more
TL;DR: In this paper, the authors performed temporal kinome analysis on human hepatocytes infected with the Erasmus isolate of MERS-CoV with peptide kinome arrays and found that ERK/MAPK and PI3K/AKT/mTOR signaling responses were specifically modulated in response to infection in vitro throughout the course of infection.
Journal ArticleDOI
The NS5 Protein of the Virulent West Nile Virus NY99 Strain Is a Potent Antagonist of Type I Interferon-Mediated JAK-STAT Signaling
Maudry Laurent-Rolle,Elena F. Boer,Kirk J. Lubick,James B. Wolfinbarger,Aaron B. Carmody,Barry Rockx,Wen Jun Liu,Joseph Ashour,W. Lesley Shupert,Michael R. Holbrook,Michael R. Holbrook,Alan D.T. Barrett,Peter W. Mason,Peter W. Mason,Marshall E. Bloom,Adolfo García-Sastre,Alexander A. Khromykh,Sonja M. Best +17 more
TL;DR: It is reported that NS5 from the virulent NY99 strain of WNV prevented pY-STAT1 accumulation, suppressedIFN-dependent gene expression, and rescued the growth of a highly IFN-sensitive virus (Newcastle disease virus) in the presence of IFN, suggesting that this protein can function as an efficient IFN antagonist.
Journal ArticleDOI
Interferon-β and mycophenolic acid are potent inhibitors of Middle East respiratory syndrome coronavirus in cell-based assays.
Brit J. Hart,Julie Dyall,Elena Postnikova,Huanying Zhou,Jason Kindrachuk,Reed F. Johnson,Gene G. Olinger,Matthew B. Frieman,Michael R. Holbrook,Peter B. Jahrling,Lisa E. Hensley +10 more
TL;DR: Compared with different IFN products, IFN-β, MPA or a combination of the two may be beneficial in the treatment of MERS-CoV or as a post-exposure intervention in high-risk patients with known exposures to MSPV.

Possible sexual transmission of ebola virus — Liberia, 2015
Athalia Christie,Gloria Davies-Wayne,Thierry Cordier-Lasalle,David J. Blackley,A. Scott Laney,Desmond E. Williams,Shivam A. Shinde,Moses Badio,Terrence Lo,Suzanne Mate,Jason T. Ladner,Michael R. Wiley,Jeffrey R. Kugelman,Gustavo Palacios,Michael R. Holbrook,Krisztina Janosko,Emmie de Wit,Neeltje van Doremalen,Vincent J. Munster,James Pettitt,Randal J. Schoepp,Leen Verhenne,Iro Evlampidou,Karsor Kollie,Sonpon B. Sieh,Alex Gasasira,Fatorma K. Bolay,Francis Kateh,Tolbert Nyenswah,Kevin M. De Cock +29 more
TL;DR: CDC now recommends that contact with semen from male Ebola survivors be avoided until more information regarding the duration and infectiousness of viral shedding in body fluids is known, and if male survivors have sex, a condom should be used correctly and consistently every time.
