Atherosclerosis, the principal cause of heart attack, stroke and gangrene of the extremities, is responsible for 50% of all mortality in the USA, Europe and Japan. The lesions result from an excessive, inflammatory-fibroproliferative response to various forms of insult to the endothelium and smooth muscle of the artery wall. A large number of growth factors, cytokines and vasoregulatory molecules participate in this process. Our ability to control the expression of genes encoding these molecules and to target specific cell types provides opportunities to develop new diagnostic and therapeutic agents to induce the regression of the lesions and, possibly, to prevent their formation.

Prevalence and incidence of diabetic microangiopathy in Japan

Recent investigations have indicated the involvement of many cytokines in cholesterol metabolism. Tumor necrosis factor (TNF)/cachectin elevates plasma very low density lipoproteins (VLDL) concentrations. In addition, granulocyte-monocyte colony-stimulating factor (GM-CSF) was reported to decrease plasma cholesterol levels. ' The similar plasma cholesterol lowering activity was also demonstrated in monocyte-colony stimulating factor (M-CSF).' On the other hand, foam cells observed in atheromatous fatty streaks are derived from monocytes/macrophages through interaction of plasma lipoproteins. We therefore attempted to investigate the effects of M-CSF on the lipoprotein metabolism of macrophages. M-CSF were obtained from Morinaga Milk Inc. (Tokyo, Japan).3 Human monocytes were prepared from blood of healthy volunteers and cultured for 10 days with and without M-CSF, and used as macrophages. Thereafter, the cholesterol ester formation from [ I4C]oleate and the cellular uptake and degradation of 1251-labeled lipoproteins were measured. Since the cellular DNA contents were hardly affected by M-CSF, the values were indicated as normalized for cellular DNA. Cholesterol esterification was stimulated in the presence of LDL or acetyl-LDL. M-CSF enhanced the stimulation by both lipoproteins. The degree of the enhancement varied greatly among different preparations of macrophages. This might be due to the variation of the responsiveness of macrophages to M-CSF. On average, the cholesterol esterification stimulated by 10 pglml of acetyl-LDL was enhanced by 24-fold as shown in TABLE 1. This enhancement of cholesterol esterification might be mainly caused by the increased uptake of 1251-acetyl-LDL, because M-CSF enhanced the uptake and degradation of "'I-acetyl-LDL by 7.5-fold (TABLE 2). However, the values for the cholesterol esterification were always greater than those for the degradation. This suggests that M-CSF increase not only the uptake of the lipoproteins but also the efficiency of esterification of cholesterol derived from the lipoproteins hydrolyzed in lysosome. This direct action of M-CSF on acyl-CoA:acyltransferase needs further investigation. As for LDL, the cholesterol esterification exceeded the degradation also (TABLES I and 2). This might be because M-CSF directly modulate the esterifying enzyme as well as the uptake of LDL as in the case of acetyl-LDL. However, the addition of butylated hydroxytoluene (20 pM) to the incubation medium decreased the uptake of '"1-LDL.

Effect of Monocyte Colony‐Stimulating Factor (M‐CSF) on Lipoprotein Metabolism

and a group of six subspecies, named the third biovariant complex (2). They are all widely distributed in soil, dust, and water and are frequently found in sputum and saliva from healthy subjects. Human pathogenicity mostly involves M. jortuitum and M. chelonae. About 50-60% of the reported infections are soft tissue abscesses or wound infections after surgery or traumatisms. Noncavitating pneumonia, endocarditis, lymphadenitis, osteomyelitis, keratitis, and fatal disseminated infections (in immunocompromised hosts) have also been reported. Cutaneous infections may occur after injections or the use of contaminated electromyography or mesotherapy needles (3).
