Molecular mechanisms for the activity of PX-478, an antitumor inhibitor of the hypoxia-inducible factor-1α
Mei Yee Koh,Taly R. Spivak-Kroizman,Sara Venturini,Sarah J. Welsh,Ryan R. Williams,D. Lynn Kirkpatrick,Garth Powis +6 more
TL;DR: It is concluded that PX-478 inhibits Hif-1α at multiple levels that together or individually may contribute to its antitumor activity against HIF-1 α-expressing tumors.
Journal ArticleDOI
The selective hypoxia inducible factor-1 inhibitor PX-478 provides in vivo radiosensitization through tumor stromal effects.
David L. Schwartz,Garth Powis,Arun Thitaikumar,Yi He,James A. Bankson,Ryan R. Williams,Robert Lemos,Junghwan Oh,Andrei Volgin,Suren Soghomonyan,Ryuichi Nishii,Mian M. Alauddin,Uday Mukhopadhay,Zhenghong Peng,William G. Bornmann,Juri G. Gelovani +15 more
TL;DR: It is shown that PX-478 prevents postradiation HIF-1 signaling and abrogates downstream stromal adaptation in C6 and HN5 reporter xenografts as measured by serial ultrasound, vascular magnetic resonance imaging, and hypoxia response element–specific micro–positron emission tomography imaging.
Journal ArticleDOI
Treatment with HIF-1α Antagonist PX-478 Inhibits Progression and Spread of Orthotopic Human Small Cell Lung Cancer and Lung Adenocarcinoma in Mice
Jörg J. Jacoby,Baruch Erez,Maria V. Korshunova,Ryan R. Williams,Kazuhisa Furutani,Osamu Takahashi,Lynn Kirkpatrick,Scott M. Lippman,Garth Powis,Michael S. O'Reilly,Roy S. Herbst +10 more
TL;DR: It is demonstrated that the PX-478, HIF-1&agr; inhibitor, had significant antitumor activity against two orthotopic models of lung adenocarcinomas and two models of SCLC.
Journal ArticleDOI
IL-24 is Expressed During Wound Repair and Inhibits TGFα induced Migration and Proliferation of Keratinocytes
Nancy Poindexter,Ryan R. Williams,Garth Powis,Emily Y. Jen,Abigail S. Caudle,Sunil Chada,Elizabeth A. Grimm +6 more
TL;DR: In conclusion, IL‐24 is expressed during wound repair and inhibits TGFα ‐induced migration and proliferation of keratinocytes in mice and successfully reprograms these cells for wound repair.
Journal ArticleDOI
Identification of Thioredoxin-Interacting Protein 1 as a Hypoxia-Inducible Factor 1α-Induced Gene in Pancreatic Cancer
Amanda F. Baker,Mei Yee Koh,Ryan R. Williams,Brian P. James,Huamin Wang,Wendy R. Tate,Alfred Gallegos,Daniel Von Hoff,Daniel Von Hoff,Haiyong Han,Garth Powis +10 more
TL;DR: Thioredoxin-interacting protein, a putative tumor suppressor gene, is induced in response to hypoxia in a HIF-1&agr;-dependent manner in pancreatic cancer cells, resulting in increased apoptosis and increased sensitivity to platinum anticancer therapy.
