Journal ArticleDOI
Fluorinated carbohydrates as lectin ligands: versatile sensors in 19F-detected saturation transfer difference NMR spectroscopy
Tammo Diercks,João P. Ribeiro,F. Javier Cañada,Sabine André,Jesús Jiménez-Barbero,Hans-Joachim Gabius +5 more
TL;DR: As a novel approach for studying carbohydrate-lectin interactions spectroscopically, the resolution and specificity of (19)F-detected NMR spectroscopy with the sensitivity of the saturation transfer difference (STD) technique are combined.
Journal ArticleDOI
Structural Basis of the Oncogenic Interaction of Phosphatase PRL-1 with the Magnesium Transporter CNNM2
Paula Giménez-Mascarell,Iker Oyenarte,Serge Hardy,Tilman Breiderhoff,Marchel Stuiver,Elie Kostantin,Tammo Diercks,Angel L. Pey,June Ereño-Orbea,María L. Martínez-Chantar,Reham Khalaf-Nazzal,Felix Claverie-Martin,Dominik N. Müller,Michel L. Tremblay,Luis Alfonso Martínez-Cruz +14 more
TL;DR: The crystal structure of PRL-1 in complex with the Bateman module of CNNM2 (CNNM2BAT), which consists of two cystathionine β-synthase (CBS) domains and represents an intracellular regulatory module of the transporter, sheds new light on the structural basis underlying the interaction between PRL phosphatases and CNNM transporters.
Journal ArticleDOI
Nucleotide binding triggers a conformational change of the CBS module of the magnesium transporter CNNM2 from a twisted towards a flat structure
María Angeles Corral-Rodríguez,Marchel Stuiver,Guillermo Abascal-Palacios,Tammo Diercks,Iker Oyenarte,June Ereño-Orbea,Alain Ibáñez de Opakua,Francisco J. Blanco,José Antonio Encinar,Vojtêch Spiwok,Hiroyuki Terashima,Alessio Accardi,Dominik Müller,Luis Alfonso Martínez-Cruz +13 more
TL;DR: The results of the present study suggest that the T568I mutation exerts its pathogenic effect in humans by constraining the conformational equilibrium of the CBS module of CNNM2, which becomes 'locked' in its flat form.
Journal ArticleDOI
Structural characterization of the N-linked glycans in the receptor binding domain of the SARS-CoV-2 spike protein and their interactions with human lectins using NMR spectroscopy.
Maria Pia Lenza,Iker Oyenarte,Tammo Diercks,Jon I. Quintana,Ana Gimeno,Alexandre Bosch,Helena Coelho,Ana Diniz,Francesca Peccati,Sandra Delgado,Mikel Valle,Oscar Millet,Nicola G. A. Abrescia,Asis Palazon,Filipa Marcelo,Gonzalo Jiménez-Osés,Jesús Jiménez-Barbero,Ana Ardá,June Ereño-Orbea +18 more
TL;DR: 3D models of the interacting complexes have been proposed based on the analysis of glycan structures of the receptor binding domain of the SARS-CoV2 spike glycoprotein expressed in human HEK293F cells by using NMR.
Journal ArticleDOI
Fluorinated Carbohydrates as Lectin Ligands: Synthesis of OH/F-Substituted N-Glycan Core Trimannoside and Epitope Mapping by 2D STD-TOCSYreF NMR spectroscopy
Tammo Diercks,Angela Simona Infantino,Luca Unione,Jesús Jiménez-Barbero,Stefan Oscarson,Hans-Joachim Gabius +5 more
TL;DR: A new 2D STD-TOCSYreF experiment is presented here that enables comprehensive epitope mapping of fluorinated glycans by combining the spectral resolution of 19 F with the spatial resolution and coverage of 1 H.
