Matrix metalloproteinases: contribution to pathogenesis, diagnosis and treatment of periodontal inflammation.
Timo Sorsa,Leo Tjäderhane,Yrjö T. Konttinen,Anneli Lauhio,Tuula Salo,Hsi-Ming Lee,Lorne M. Golub,David L. Brown,Päivi Mäntylä +8 more
TL;DR: MMP‐inhibitor (MMPI)‐drugs, such as doxycycline, can be used as adjunctive medication to augment both the scaling and root planing‐treatment of periodontitis locally and to reduce inflammation systematically.
Journal ArticleDOI
Tetracyclines Inhibit Connective Tissue Breakdown by Multiple Non-Antimicrobial Mechanisms
Lorne M. Golub,Hsi-Ming Lee,Maria Emanuel Ryan,William V. Giannobile,Jeffrey B. Payne,Timo Sorsa +5 more
TL;DR: Longitudinal double-blind studies on humans with adult periodontitis have demonstrated that a sub-antimicrobial dose of doxycycline (previously reported to suppress collagenase activity in the periodontal pocket) is safe and effective and has been approved by the FDA as an adjunct to scaling and root planing.
Journal ArticleDOI
Tumor targeting with a selective gelatinase inhibitor
Erkki Koivunen,Wadih Arap,Heli Valtanen,Aija Rainisalo,Oula Penate Medina,Pia Heikkilä,Carmela Kantor,Carl G. Gahmberg,Tuula Salo,Yrjö T. Konttinen,Timo Sorsa,Erkki Ruoslahti,Renata Pasqualini +12 more
TL;DR: It is shown that cyclic peptides containing the sequence HWGF are potent and selective inhibitors of M MP-2 and MMP-9 but not of several other MMP family members, and its prototype synthetic peptide, CTTHWGFTLC, inhibits the migration of human endothelial cells and tumor cells.
Journal ArticleDOI
Matrix metalloproteinases (MMPs) in oral diseases.
TL;DR: The history, the current status and the future aspects of the work mainly of the research groups looking at the presence and activity of various MMPs in different oral diseases are described, as well as some of the MMP-related aspects that may facilitate the development of new means of diagnosis and treatment of oral diseases.
Journal ArticleDOI
The Activation and Function of Host Matrix Metalloproteinases in Dentin Matrix Breakdown in Caries Lesions
TL;DR: Results demonstrate the pH-dependent activation mechanism of MMPs, which may have a distinct role in different physiological and pathological conditions and demonstrate that host M MPs, activated by bacterial acids, have a crucial role in the destruction of dentin by caries.
