Journal ArticleDOI
Tracking the origins and drivers of subclonal metastatic expansion in prostate cancer
Matthew K.H. Hong,Geoff Macintyre,David C. Wedge,Peter Van Loo,Keval M. Patel,Sebastian Lunke,Ludmil B. Alexandrov,Clare Sloggett,Marek Cmero,Francesco Marass,Dana W.Y. Tsui,Stefano Mangiola,Andrew Lonie,Haroon Naeem,Nikhil Sapre,Pramit M. Phal,Natalie Kurganovs,Xiaowen Chin,Michael Kerger,Anne Y. Warren,David E. Neal,Vincent Gnanapragasam,Nitzan Rosenfeld,John Pedersen,Andrew Ryan,Izhak Haviv,Anthony J. Costello,Niall M. Corcoran,Christopher M. Hovens +28 more
TL;DR: The precise direction of metastatic spread is revealed across four lethal prostate cancer patients using whole-genome and ultra-deep targeted sequencing of longitudinally collected primary and metastatic tumours and analysis of mutations associated with metastasis reveals an enrichment of TP53 mutations.
Journal ArticleDOI
Levels of a subpopulation of platelets, but not circulating endothelial cells, predict early treatment failure in prostate cancer patients after prostatectomy
C K E Wong,Benjamin Namdarian,J. Chua,Xiaowen Chin,R Speirs,Thanh T. Nguyen,M Fankhauser,John Pedersen,Anthony J. Costello,Anthony J. Costello,Niall M. Corcoran,Niall M. Corcoran,Christopher M. Hovens,Christopher M. Hovens +13 more
TL;DR: Elevated levels of a distinct subpopulation of circulating platelets were an independent predictor for early biochemical recurrence in prostate cancer patients within the first year from prostatectomy.
Journal ArticleDOI
Loss of APKC expression independently predicts tumor recurrence in superficial bladder cancers.
Benjamin Namdarian,Edwin Ck Wong,Ryan C. Galea,John Pedersen,Xiaowen Chin,Robert Speirs,Patrick O. Humbert,Anthony J. Costello,Niall M. Corcoran,Christopher M. Hovens +9 more
TL;DR: Investigation of the role of tight junction proteins, Par3/Par6/atypical protein kinase C, Discs large, and Scribble in human bladder pathogenesis found loss of APKC expression in superficial bladder tumors is a strong predictor of tumor recurrence.
Journal ArticleDOI
Dynamin II function is required for EGF-mediated Stat3 activation but not Erk1/2 phosphorylation
TL;DR: Data suggest that the activation status of Stat3 and Erk1/2 and the sustainability of these signals are potentially due to the spatial and temporal control of the EGFR within the cell, which may have implications on therapeutic targeting and efficacy when using inhibitors to proteins either regulating endocytosis and/or signalling.
Journal ArticleDOI
Percutaneous image-guided biopsy of prostate cancer metastases yields samples suitable for genomics and personalised oncology
Matthew K.H. Hong,Nikhil Sapre,Pramit M. Phal,Geoff Macintyre,Xiaowen Chin,John Pedersen,Andrew Ryan,Michael Kerger,Anthony J. Costello,Niall M. Corcoran,Christopher M. Hovens +10 more
TL;DR: The procurement of osseous metastatic prostate cancer from live patients, including the use of image-guided biopsy, is safe and feasible and sufficient tissue can be obtained in a manner such that extracted nucleic acids are suitable for genomics research.
