Yi Lynn Liang,Maryam Khoshouei,Mazdak Radjainia,Yan Zhang,Alisa Glukhova,Jeffrey T. Tarrasch,David M. Thal,Sebastian G.B. Furness,George Christopoulos,Thomas Coudrat,Radostin Danev,Wolfgang Baumeister,Laurence J. Miller,Arthur Christopoulos,Brian K. Kobilka,Denise Wootten,Georgios Skiniotis,Patrick M. Sexton +17 more
TL;DR: The structure of a full-length class B receptor, the calcitonin receptor, in complex with peptide ligand and heterotrimeric Gαsβγ protein determined by Volta phase-plate single-particle cryo-electron microscopy provides a new framework for understanding G-protein-coupled receptor function.
Journal ArticleDOI
Structure of the adenosine-bound human adenosine A 1 receptor-G i complex.
Christopher J Draper-Joyce,Maryam Khoshouei,Maryam Khoshouei,David M. Thal,Yi Lynn Liang,Anh Nguyen,Sebastian G.B. Furness,Hariprasad Venugopal,Jo-Anne Baltos,Jürgen M. Plitzko,Radostin Danev,Wolfgang Baumeister,Lauren T. May,Denise Wootten,Denise Wootten,Patrick M. Sexton,Patrick M. Sexton,Alisa Glukhova,Arthur Christopoulos +18 more
TL;DR: The cryo-electron microscopy structure of the humanAdenosine A1 receptor in complex with adenosine and heterotrimeric Gi2 protein provides molecular insights into receptor and G-protein selectivity.
Journal ArticleDOI
Phase-plate cryo-EM structure of a biased agonist-bound human GLP-1 receptor-Gs complex.
Yi Lynn Liang,Maryam Khoshouei,Alisa Glukhova,Sebastian G.B. Furness,Peishen Zhao,Lachlan Clydesdale,Cassandra Koole,Tin T. Truong,David M. Thal,Saifei Lei,Mazdak Radjainia,Radostin Danev,Wolfgang Baumeister,Ming-Wei Wang,Ming-Wei Wang,Laurence J. Miller,Laurence J. Miller,Arthur Christopoulos,Patrick M. Sexton,Patrick M. Sexton,Denise Wootten +20 more
TL;DR: The structure of the human GLP-1 receptor in complex with the G protein-biased peptide exendin-P5 and a Gαs heterotrimer is described and provides insights into the molecular basis of biased agonism.
Journal ArticleDOI
Cryo-EM structure of the active, G s -protein complexed, human CGRP receptor
Yi Lynn Liang,Maryam Khoshouei,Maryam Khoshouei,Giuseppe Deganutti,Alisa Glukhova,Cassandra Koole,Thomas S. Peat,Mazdak Radjainia,Mazdak Radjainia,Jürgen M. Plitzko,Wolfgang Baumeister,Laurence J. Miller,Laurence J. Miller,Debbie L. Hay,Arthur Christopoulos,Christopher A. Reynolds,Denise Wootten,Denise Wootten,Patrick M. Sexton,Patrick M. Sexton +19 more
TL;DR: Structural insight is provided into the regulation of G-protein-coupled receptors by receptor activity modifying protein 1 and molecular dynamics simulations indicate that RAMP1 provides stability to the receptor complex, particularly in the positioning of the extracellular domain of CLR.
Journal ArticleDOI
Ligand-dependent modulation of G protein conformation alters drug efficacy
Sebastian G.B. Furness,Yi Lynn Liang,Cameron J. Nowell,Michelle L. Halls,Peter J. Wookey,Emma Dal Maso,Asuka Inoue,Asuka Inoue,Arthur Christopoulos,Denise Wootten,Patrick M. Sexton +10 more
TL;DR: It is reported that ligand efficacy at the calcitonin GPCR (CTR) is also correlated with ligand-dependent alterations to G protein conformation, demonstrating that factors influencing efficacy extend beyond receptor conformation(s) and expands understanding of the molecular basis for how G proteins control/influence efficacy.
