Journal ArticleDOI
Rational design of true monomeric and bright photoactivatable fluorescent proteins
Mingshu Zhang,Hao Chang,Hao Chang,Yongdeng Zhang,Yongdeng Zhang,Junwei Yu,Junwei Yu,Lijie Wu,Wei Ji,Juanjuan Chen,Bei Liu,Bei Liu,Jingze Lu,Yingfang Liu,Jun-Long Zhang,Pingyong Xu,Tao Xu,Tao Xu +17 more
TL;DR: It is shown that mEos2 forms oligomers at high concentrations and forms aggregates when labeling membrane proteins, limiting its application as a fusion partner.
Journal ArticleDOI
Ultra-High Resolution 3D Imaging of Whole Cells
Fang Huang,Fang Huang,George Sirinakis,George Sirinakis,Edward S. Allgeyer,Edward S. Allgeyer,Lena K. Schroeder,Whitney C. Duim,Whitney C. Duim,Emil B. Kromann,Thomy Phan,Felix Rivera-Molina,Jordan R. Myers,Irnov Irnov,Mark D. Lessard,Yongdeng Zhang,Mary Ann Handel,Christine Jacobs-Wagner,C. Patrick Lusk,James E. Rothman,Derek Toomre,Martin J. Booth,Joerg Bewersdorf +22 more
TL;DR: The development of whole-cell 4Pi single-molecule switching nanoscopy (W-4PiSMSN), an optical nanoscope that allows imaging of three-dimensional structures at 10- to 20-nm resolution throughout entire mammalian cells, is presented.
Journal ArticleDOI
Long time-lapse nanoscopy with spontaneously blinking membrane probes
Hideo Takakura,Yongdeng Zhang,Roman S. Erdmann,Alexander D. Thompson,Yu Lin,Brian McNellis,Felix Rivera-Molina,Shin-nosuke Uno,Mako Kamiya,Mako Kamiya,Yasuteru Urano,Yasuteru Urano,James E. Rothman,Joerg Bewersdorf,Alanna Schepartz,Derek Toomre +15 more
TL;DR: A set of high-density, environment-sensitive (HIDE) membrane probes, based on the membrane-permeable silicon-rhodamine dye HMSiR, that assemble in situ and enable long time-lapse, live-cell nanoscopy of discrete cellular structures and organelles with high spatiotemporal resolution are introduced.
Journal ArticleDOI
A unique series of reversibly switchable fluorescent proteins with beneficial properties for various applications
Hao Chang,Mingshu Zhang,Wei Ji,Juanjuan Chen,Yongdeng Zhang,Bei Liu,Jingze Lu,Jun-Long Zhang,Pingyong Xu,Tao Xu +9 more
TL;DR: A series of monomeric green RSFPs with beneficial optical characteristics such as high photon output per switch, high photostability, a broad range of switching rate, and pH-dependence are developed, which make them potentially useful for various applications.
Journal ArticleDOI
Dynamic nanoscale morphology of the ER surveyed by STED microscopy.
Lena K. Schroeder,Andrew E.S. Barentine,Holly Merta,Sarah Schweighofer,Yongdeng Zhang,David Baddeley,David Baddeley,Joerg Bewersdorf,Shirin Bahmanyar +8 more
TL;DR: The first quantitative analysis of ER membrane structure and dynamics at the nanoscale is provided, revealing that the ER in living cells is not limited to uniform sheets and tubules and suggests the ER contains a continuum of membrane structures that includes dynamic nanoholes in sheets as well as clustered tubules.
