Watson,Jack Baty,Jennifer Ivanovich,Sharon Heath,William D. Shannon,Rakesh Nagarajan,Matthew J. Walter,Daniel C. Link,Timothy A. Graubert,John F. DiPersio,Richard K. Wilson +47 more
TL;DR: This study establishes whole-genome sequencing as an unbiased method for discovering cancer-initiating mutations in previously unidentified genes that may respond to targeted therapies.
Journal ArticleDOI
Acquired copy number alterations in adult acute myeloid leukemia genomes
Matthew J. Walter,Jacqueline E. Payton,Rhonda E. Ries,William D. Shannon,Hrishikesh Deshmukh,Yu Zhao,Jack Baty,Sharon Heath,Peter Westervelt,Mark A. Watson,Michael H. Tomasson,Rakesh Nagarajan,Brian O’Gara,Clara D. Bloomfield,Krzysztof Mrózek,Rebecca R. Selzer,Todd Richmond,Jacob O. Kitzman,Joel Geoghegan,Peggy S. Eis,Rachel Maupin,Robert S. Fulton,Michael D. McLellan,Richard K. Wilson,Elaine R. Mardis,Daniel C. Link,Timothy A. Graubert,John F. DiPersio,Timothy J. Ley +28 more
TL;DR: The use of an unbiased high-resolution genomic screen identified many genes not previously implicated in AML that may be relevant for pathogenesis, along with many known oncogenes and tumor suppressor genes.
Journal ArticleDOI
Somatic mutations and germline sequence variants in the expressed tyrosine kinase genes of patients with de novo acute myeloid leukemia.
Michael H. Tomasson,Zhifu Xiang,Richard A. Walgren,Yu Zhao,Yumi Kasai,Tracie L. Miner,Rhonda E. Ries,Olga Y. Lubman,Daved H. Fremont,Michael D. McLellan,Jacqueline E. Payton,Peter Westervelt,John F. Ley +24 more
TL;DR: High-throughput resequencing of the kinase domains of 26 TK genes expressed in most AML patients found 4 novel somatic mutations and novel germline sequence changes encoding amino acid substitutions in 14 Tk genes, including TYK2, which had the largest number of nonsynonymous sequence variants (11 total detected).
Journal ArticleDOI
The histone demethylase LSD1/KDM1A promotes the DNA damage response
Nima Mosammaparast,Haeyoung Kim,Benoit Laurent,Yu Zhao,Hui Jun Lim,Mona C. Majid,Sebastian Dango,Yuying Luo,Kristina Hempel,Mathew E. Sowa,Steven P. Gygi,Hanno Steen,J. Wade Harper,Bruce A. Yankner,Yang Shi +14 more
TL;DR: The E3 ubiquitin ligase RNF168 recruits LSD1 to DNA damage sites, where it reduces histone methylation upstream of 53BP1 recruitment during the DNA damage response.
Journal ArticleDOI
Identification of somatic JAK1 mutations in patients with acute myeloid leukemia
Zhifu Xiang,Yu Zhao,Vesselin Mitaksov,Daved H. Fremont,Yumi Kasai,AnnaLynn Molitoris,Rhonda E. Ries,Tracie L. Miner,Michael D. McLellan,John F. DiPersio,Daniel C. Link,Jacqueline E. Payton,Timothy A. Graubert,Mark F. Watson,William D. Shannon,Sharon Heath,Rakesh Nagarajan,Elaine R. Mardis,Rick K. Wilson,Timothy J. Ley,Michael H. Tomasson +20 more
TL;DR: In this paper, the entire coding region of JAK1 was screened by total exonic resequencing of bone marrow DNA samples from 94 patients with de novo acute myeloid leukemia (AML).
