A new mutation rpoD800, affecting the sigma subunit of E. coli RNA polymerase is allelic to two other sigma mutants.

TLDR
It is argued that the single locus affected is the structural gene for the sigma subunit of E. coli RNA polymerase, which renders sigma about 10-fold more thermolabile than the wild type sigma at 45°C in vitro.
Abstract
We have characterized a new mutation rpoD800 affecting the sigma gene of E. coli. Upon transfer to high temperature, a strain with the rpoD800 mutation ceases growth within 30 min. We find that this mutation renders sigma about 10-fold more thermolabile than the wild type sigma at 45°C in vitro. We have compared the temperature profile for inactivation of wild type and mutant sigma and find that the mutant inactivates at a temperature about 9° C lower than does the wild type.

A mutant Escherichia coli σ70 subunit of RNA polymerase with altered promoter specificity

TL;DR: Results suggest that this region of sigma 70 is directly involved in recognition of the -35 hexamer, a region that is also homologous with the helix-turn-helix motif of DNA-binding proteins.

Linkage Map of Escherichia coli K-12, Edition 10: The Traditional Map

TL;DR: This map is an update of the edition 9 map by Berlyn et al. and adds new genes discovered and established since 1996 and eliminates those shown to correspond to other known genes.

RNA polymerase rifampicin resistance mutations in Escherichia coli: sequence changes and dominance.

TL;DR: Five recombinant plasmids carrying rpoB rifampicin-resistant RNA-polymerase genes were obtained and sequence analysis revealed certain structural changes in the rPoB gene which specify corresponding alterations in the β-subunit of RNA polymerase.

Factor stimulating transcription by RNA polymerase.

TL;DR: A protein component usually associated with RNA polymerase can be separated from the enzyme by chromatography on phosphocellulose, and the polymerase is unable to transcribe T4 DNA unless this factor is added back.

Evidence for the direct involvement of RNA in the initiation of DNA replication in Escherichia coli 15T.

TL;DR: It is proposed that RNA participates directly in the replication of DNA and that the proteins required for replication are assembled around an RNA core forming the replication complex.
