Microassay for cathepsin D shows an unexpected effect of cycloheximide on limb-bone rudiments in organ culture
TLDR
A simple microdetermination for the lysosomal enzyme, cathepsin D, has been developed and cycloheximide and puromycin, inhibitors of protein synthesis, immediately stopped the synthesis of new enzyme, but caused a brief burst of release of preformed enzyme from the tissue.About:

Removal of Z-lines and alpha-actinin from isolated myofibrils by a calcium-activated neutral protease.
TL;DR: A calcium-activated factor (CaAF) has been isolated and partially purified from the post-myofibrillar supernatant fraction of rabbit skeletal muscle and a protein with the properties of alpha-actinin (identical electrophoretic mobility, and ability to accelerate the Mg2+-activated ATPase of reconstituted actomyosin) was isolated from the supernatants of CaAF-treated myofibrils.

Secretion of lysosomal hydrolases by stimulated and nonstimulated macrophages.
J Schnyder,M Baggiolini +1 more
TL;DR: Lysosomal enzyme secretion in macrophages appears to depend on protein synthesis, since it was blocked by low concentrations of cycloheximide which neither affected cell viability nor lowered the intracellular enzyme levels.

Potent new inhibitors of human renin
TL;DR: High in vitro inhibitory potencies of H-113 and H-142 are believed to be due to the ability of the reduced moiety —CH2—NH— to act as a non-hydrolysable analogue of the tetrahedral transition state formed during hydrolysis of the (10–11) peptide bond.

Compositional studies of myofibrils from rabbit striated muscle.
J D Etlinger,R Zak,D A Fischman +2 more
TL;DR: The localization of high-molecular-weight proteins in the sarcomere of striated muscle has been studied by coordinated electron-microscopic and sodium dodecyl sulfate (SDS) gel electrophoretic analysis of native myofilaments and extracted and digested myofibrils.

The use of urinary N-acetyl-β-glucosaminidase in human renal toxicology I. Partial biochemical characterization and excretion in humans and release from the isolated perfused rat kidney☆
TL;DR: It is concluded that, with certain restrictions, N-acetyl-β-glucosaminidase excretion can be employed as a marker of the rate of release of at least several acid hydrolases from the human kidney.

Cathepsin D. Purification of isoenzymes from human and chicken liver.
TL;DR: A modified analytical method of isoelectric focusing in polyacrylamide gel indicated a high degree of homogeneity of the purified beta and gamma isoenzymes from each species, and this was supported by their constant high specific activities.

INTRACELLULAR TRANSPORT OF SECRETORY PROTEINS IN THE PANCREATIC EXOCRINE CELL : III. Dissociation of Intracellular Transport from Protein Synthesis
TL;DR: Transport and protein synthesis are separable processes; intracellular transport is not the result of a continuous delivery of secretory proteins from attached polysomes to the cisternae of the rough endoplasmic reticulum; and transport isNot dependent on the synthesis of "specific" nonsecretory proteins within the time limits tested.
Journal ArticleDOI
Lysosomal acid proteinase of rabbit liver
TL;DR: It is concluded that this enzyme is a form of cathepsin D, which may be widely distributed in lysosomes, and was competitively inhibited by 3-phenylpyruvic acid at low concentrations.

High-efficiency liquid-scintillation counting of 14C-labelled material in aqueous solution and determination of specific activity of labelled proteins

Endocytosis of sugars in embryonic skeletal tissues in organ culture. IV. Lysosomal and other biochemical effects. General discussion.
TL;DR: Experiments with [14C] dextran indicated that the cytoplasmic vacuolation was due not to a higher rate of endocytosis, but rather to an abnormally long persistence of the pinocytotic vacuoles.
