Journal ArticleDOI
Quantitative and sensitive detection of rare mutations using droplet-based microfluidics
Deniz Pekin,Yousr Skhiri,Jean-Christophe Baret,Jean-Christophe Baret,Delphine Le Corre,Linas Mazutis,Chaouki Ben Salem,Florian Millot,Abdeslam El Harrak,J. Brian Hutchison,Jonathan W. Larson,Darren R. Link,Pierre Laurent-Puig,Andrew D. Griffiths,Valérie Taly +14 more
TL;DR: The technique enabled the determination of mutant allelic specific imbalance (MASI) in several cancer cell-lines and the precise quantification of a mutated KRAS gene in the presence of a 200,000-fold excess of unmutated KRAS genes.
Journal ArticleDOI
High-resolution dose-response screening using droplet-based microfluidics.
Oliver J. Miller,Abdeslam El Harrak,Thomas Mangeat,Jean-Christophe Baret,Lucas Frenz,Bachir El Debs,Estelle Mayot,Michael L. Samuels,Eamonn K. Rooney,Pierre Dieu,Martin Galvan,Darren R. Link,Andrew D. Griffiths +12 more
TL;DR: A robust microfluidic approach that increases the number of data points to approximately 10,000 per compound is described, which reveals complex dose–response relationships unambiguously and results in IC50 values that are highly precise and reproducible.
Journal ArticleDOI
Droplet‐Based Microreactors for the Synthesis of Magnetic Iron Oxide Nanoparticles
Lucas Frenz,Abdeslam El Harrak,Matthias Pauly,Sylvie Begin-Colin,Andrew D. Griffiths,Jean-Christophe Baret +5 more
Journal ArticleDOI
Kinetic aspects of emulsion stabilization by surfactants: a microfluidic analysis.
TL;DR: It is demonstrated that, in addition to the classical effect of stabilization by an increase of surfactant concentration, the dynamics of adsorption of Surfactant at the water-oil interface is a key element for droplet stabilization.
Journal ArticleDOI
Enhanced chemical synthesis at soft interfaces: a universal reaction-adsorption mechanism in microcompartments
Ali Fallah-Araghi,Kamel Meguellati,Jean-Christophe Baret,Abdeslam El Harrak,Thomas Mangeat,Martin Karplus,Martin Karplus,Sylvain Ladame,Sylvain Ladame,Carlos M. Marques,Andrew D. Griffiths,Andrew D. Griffiths +11 more
TL;DR: Reaction thermodynamics is modified by compartmentalization at the mesoscale--without confinement on the molecular scale--leading to a universal mechanism for improving unfavorable reactions.
