Journal ArticleDOI
The acute promyelocytic leukemia-specific PML-RARα fusion protein inhibits differentiation and promotes survival of myeloid precursor cells
Francesco Grignani,Pier Francesco Ferrucci,Ugo Testa,Giampaolo Talamo,Marta Fagioli,Myriam Alcalay,Amedea Mencarelli,Fausto Grignani,Cesare Peschle,Cesare Peschle,Ildo Nicoletti,Pier Giuseppe Pelicci +11 more
TL;DR: Expressing the PML-RAR alpha protein in U937 myeloid precursor cells showed that they lost the capacity to differentiate under the action of different stimuli, acquired enhanced sensitivity to retinoic acid, and exhibited a higher growth rate consequent to diminished apoptotic cell death.
Journal Article
High-Efficiency Gene Transfer and Selection of Human Hematopoietic Progenitor Cells with a Hybrid EBV/Retroviral Vector Expressing the Green Fluorescence Protein
Francesco Grignani,Todd Kinsella,Amedea Mencarelli,Mauro Valtieri,Daniela Riganelli,Fausto Grignani,Luisa Lanfrancone,Cesare Peschle,Garry P. Nolan,Pier Giuseppe Pelicci +9 more
TL;DR: The high viral titer and the easy obtainment of homogeneously infected cell populations without drug selection procedures make PINCO an ideal vector for gene transfer of human primary hemopoietic cells.
Journal Article
Heterogeneous nuclear expression of the promyelocytic leukemia (PML) protein in normal and neoplastic human tissues.
Marcello Gambacorta,L. Flenghi,Marta Fagioli,Stefano Pileri,Lorenzo Leoncini,Barbara Bigerna,Roberta Pacini,Laura Natali Tanci,Laura Pasqualucci,Stefano Ascani,Amedea Mencarelli,Arcangelo Liso,Pier Giuseppe Pelicci,Brunangelo Falini +13 more
TL;DR: It is found that PML is frequently overexpressed in Hodgkin and Reed-Sternberg cells of Hodgkin's disease, a tumor of cytokine-producing cells, and among solid tumors, overexpression of PML was frequently found in carcinomas of larynx and thyroid (papillary), epithelial thymomas, and Kaposi's sarcoma.
Journal ArticleDOI
Cooperation between the RING + B1-B2 and coiled-coil domains of PML is necessary for its effects on cell survival
Marta Fagioli,Myriam Alcalay,Lucia Tomassoni,Pier Francesco Ferrucci,Amedea Mencarelli,Daniela Riganelli,Francesco Grignani,Tullio Pozzan,Ildo Nicoletti,Fausto Grignani,Pier Giuseppe Pelicci,Pier Giuseppe Pelicci +11 more
TL;DR: Results indicate that PML is involved in cell survival regulation and that the PML component of the fusion protein retains intact biological activity, thereby suggesting that the effects of PML/RARα on survival derive from the activation of the incorporated PML sequence.
Journal ArticleDOI
Natural killer cell-mediated lysis of autologous cells modified by gene therapy.
Concetta Liberatore,Marusca Capanni,Nicola Albi,Isabella Volpi,Elena Urbani,Loredana Ruggeri,Amedea Mencarelli,Francesco Grignani,Andrea Velardi +8 more
TL;DR: It is demonstrated that NK cells can selectively detect the expression of heterologous genes, and this observations provide a general model of the NK cell–mediated control of viral infections.
