Journal ArticleDOI
Tumour evolution inferred by single-cell sequencing
Nicholas Navin,Jude Kendall,Jennifer Troge,Peter Andrews,Linda Rodgers,Jeanne McIndoo,Kerry Cook,Asya Stepansky,Dan Levy,Diane Esposito,Lakshmi Muthuswamy,Alexander Krasnitz,W. Richard McCombie,James W. Hicks,Michael Wigler +14 more
TL;DR: It is shown that with flow-sorted nuclei, whole genome amplification and next generation sequencing the authors can accurately quantify genomic copy number within an individual nucleus and indicate that tumours grow by punctuated clonal expansions with few persistent intermediates.
Journal ArticleDOI
De Novo Gene Disruptions in Children on the Autistic Spectrum
Ivan Iossifov,Michael Ronemus,Dan Levy,Zihua Wang,Inessa Hakker,Julie Rosenbaum,Boris Yamrom,Yoon-ha Lee,Giuseppe Narzisi,Anthony Leotta,Jude Kendall,Ewa A. Grabowska,Beicong Ma,Steven Marks,Linda Rodgers,Asya Stepansky,Jennifer Troge,Peter Andrews,Mitchell A. Bekritsky,Kith Pradhan,Elena Ghiban,Melissa Kramer,Jennifer Parla,Ryan Demeter,Lucinda Fulton,Robert S. Fulton,Vincent Magrini,Kenny Ye,Jennifer C. Darnell,Robert B. Darnell,Robert B. Darnell,Elaine R. Mardis,Richard K. Wilson,Michael C. Schatz,Richard W. McCombie,Michael Wigler +35 more
TL;DR: Exome sequencing of 343 families, each with a single child on the autism spectrum and at least one unaffected sibling, reveals de novo small indels and point substitutions, which suggest FMRP-associated genes are especially dosage-sensitive targets of cognitive disorders.
Journal ArticleDOI
Genome-wide copy number analysis of single cells.
Timour Baslan,Jude Kendall,Linda Rodgers,Hilary Cox,Michael Riggs,Asya Stepansky,Jennifer Troge,Kandasamy Ravi,Diane Esposito,B. Lakshmi,Michael Wigler,Nicholas Navin,James W. Hicks +12 more
TL;DR: A protocol that allows for the genome-wide copy number analysis of single nuclei isolated from mixed populations of cells is presented and informatic approaches that correct for biases inherent in the WGA procedure and allow for accurate determination of copy number profiles are outlined.
Journal ArticleDOI
A RSC/Nucleosome Complex Determines Chromatin Architecture and Facilitates Activator Binding
Monique Floer,Xin Wang,Vidya Prabhu,Georgina Berrozpe,Santosh Narayan,Dan Spagna,David Alvarez,Jude Kendall,Alexander Krasnitz,Asya Stepansky,James W. Hicks,Gene O. Bryant,Mark Ptashne +12 more
TL;DR: The results show that both prior to and following induction, specific DNA-binding proteins are the predominant determinants of chromatin architecture at the GAL1/10 genes.
Journal ArticleDOI
Rapid phenotypic and genomic change in response to therapeutic pressure in prostate cancer inferred by high content analysis of single circulating tumor cells.
Angel E. Dago,Asya Stepansky,Anders Carlsson,Madelyn Luttgen,Jude Kendall,Timour Baslan,Anand Kolatkar,Michael Wigler,Kelly Bethel,Mitchell E. Gross,James W. Hicks,Peter Kuhn +11 more
TL;DR: The timeframe of tumor evolution in response to therapy is demonstrated and a framework for the multi-scale analysis of fluid biopsies to quantify and monitor disease evolution in individual patients is provided.
