Journal ArticleDOI
Oxidative damage is the earliest event in Alzheimer disease.
Akihiko Nunomura,George Perry,Gjumrakch Aliev,Keisuke Hirai,Keisuke Hirai,Atsushi Takeda,Elizabeth Balraj,Paul K. Jones,Hossein A. Ghanbari,Takafumi Wataya,Shun Shimohama,Shigeru Chiba,Craig S. Atwood,Robert B. Petersen,Mark A. Smith +14 more
TL;DR: The observations indicate that increased oxidative damage is an early event in AD that decreases with disease progression and lesion formation and suggest that AD is associated with compensatory changes that reduce damage from reactive oxygen.
Journal ArticleDOI
Genome-wide association study identifies common variants at four loci as genetic risk factors for Parkinson's disease
Wataru Satake,Yuko Nakabayashi,Yuko Nakabayashi,Ikuko Mizuta,Ikuko Mizuta,Yushi Hirota,Yushi Hirota,Chiyomi Ito,Chiyomi Ito,Michiaki Kubo,Takahisa Kawaguchi,Tatsuhiko Tsunoda,Masahiko Watanabe,Atsushi Takeda,Hiroyuki Tomiyama,Kenji Nakashima,Kazuko Hasegawa,Fumiya Obata,Takeo Yoshikawa,Hideshi Kawakami,Saburo Sakoda,Mitsutoshi Yamamoto,Nobutaka Hattori,Miho Murata,Yusuke Nakamura,Tatsushi Toda,Tatsushi Toda +26 more
TL;DR: The results identify two new PD susceptibility loci, show involvement of autosomal dominant parkinsonism loci in typical PD and suggest that population differences contribute to genetic heterogeneity in PD.
Journal ArticleDOI
Role of mitochondrial dysfunction in Alzheimer's disease.
Rudy J. Castellani,Keisuke Hirai,Gjumrakch Aliev,Kelly L. Drew,Akihiko Nunomura,Atsushi Takeda,Adam D. Cash,Mark E. Obrenovich,George Perry,Mark A. Smith +9 more
TL;DR: The causes and consequences of mitochondrial disturbances in Alzheimer's disease as well as how this information might impact on therapeutic approaches to this disease are reviewed.
Journal ArticleDOI
Differential activation of neuronal ERK, JNK/SAPK and p38 in Alzheimer disease: the 'two hit' hypothesis.
Xiongwei Zhu,Rudolph J. Castellani,Atsushi Takeda,Akihiko Nunomura,Craig S. Atwood,George Perry,Mark A. Smith +6 more
TL;DR: Findings indicate that MAPK pathways are differentially activated during the course of Alzheimer disease and suggest that both oxidative stress and abnormalities in mitosis can independently serve to initiate, but both are necessary to propagate, disease pathogenesis.
Journal ArticleDOI
Is oxidative damage the fundamental pathogenic mechanism of Alzheimer's and other neurodegenerative diseases?
George Perry,Akihiko Nunomura,Akihiko Nunomura,Keisuke Hirai,Keisuke Hirai,Xiongwei Zhu,Mar Prez,Jess Avila,Rudolph J. Castellani,Craig S. Atwood,Gjumrakch Aliev,Lawrence M. Sayre,Atsushi Takeda,Atsushi Takeda,Mark A. Smith +14 more
TL;DR: The types of oxidative damage seen in AD, sites involved, possible origin, relationship to lesions, and compensatory changes are reviewed, and other neurodegenerative diseases where oxidative stress has been implicated are considered.
