Brugge,Aviv Regev,Gad Getz,Eytan Ruppin,Yosef Yarden +38 more
TL;DR: Treatment of lung cancer with EGFR inhibitors with AXL overexpression cured patient-derived xenografts and uncovered another hypermutator, AXL-driven activation of MYC and increased purine synthesis that is unbalanced by pyrimidines.
Journal ArticleDOI
RAC1 Alterations Induce Acquired Dabrafenib Resistance in Association with Anaplastic Transformation in a Papillary Thyroid Cancer Patient
Rozita Bagheri-Yarmand,Naifa L. Busaidy,Elena McBeath,Brian P. Danysh,Kurt W. Evans,Tyler J. Moss,Argun Akcakanat,Patrick Kwok Shing Ng,Christina M. Knippler,Jalyn A. Golden,Michelle D. Williams,Asha S. Multani,Maria E. Cabanillas,Kenna R. Shaw,Funda Meric-Bernstam,Manisha H. Shah,Matthew D. Ringel,Marie Claude Hofmann +17 more
TL;DR: In this article, a new RAC1 (P34R) mutation acquired during dabrafenib treatment in a progressive metastatic lesion with ATC phenotype was identified.

Brown,Laxmi Parida,Stephan Stilgenbauer,Gad Getz,Catherine J. Wu +53 more
TL;DR: This study defines RS-specific alterations and provides a molecular definition of RS, identifies distinct genetic subtypes of RS with prognostic significance, traces the evolutionary path to RS and suggests future strategies for improved detection.
Journal ArticleDOI
Pirtobrutinib targets BTK C481S in ibrutinib-resistant CLL but second-site BTK mutations lead to resistance
Aisha Naeem,Filippo Utro,Qing Wang,Justin Cha,Mauno Vihinen,Stephen P. Martindale,Yinglu Zhou,Yue-huan Ren,Svitlana Tyekucheva,Annette S. Kim,Stacey M. Fernandes,Gordon Saksena,Kahn Rhrissorrakrai,Chaya Levovitz,Brian P. Danysh,Kara Slowik,Raquel A. Jacobs,Matthew S. Davids,James A. Lederer,Rula Zain,C. I. E. Smith,Ignaty Leshchiner,Laxmi Parida,Gad Getz,Jennifer R. Brown +24 more
TL;DR: It is shown that pirtobrutinib potently inhibits BTK-mediated functions including BCR signaling, cell viability and CCL3/CCL4 chemokine production in both BTK wild-type (WT) and C481S mutant CLL cells.
