Journal ArticleDOI
Arterial Calcifications and Bone Histomorphometry in End-Stage Renal Disease
Gérard M. London,Caroline Marty,Sylvain J. Marchais,Alain P. Guerin,Fabien Metivier,Marie Christine de Vernejoul +5 more
TL;DR: It is suggested that therapeutic interventions associated with excessive lowering of parathyroid activity (parathyroidectomy, excessive calcium or aluminum load) favor lower bone turnover and adynamic bone disease, which could influence the development and progression of AC.
Journal ArticleDOI
Thrombopoietin receptor activation by myeloproliferative neoplasm associated calreticulin mutants
Ilyas Chachoua,Ilyas Chachoua,Christian Pecquet,Christian Pecquet,Mira El-Khoury,Harini Nivarthi,Roxana-Irina Albu,Roxana-Irina Albu,Caroline Marty,Vitalina Gryshkova,Vitalina Gryshkova,Jean-Philippe Defour,Jean-Philippe Defour,Gaëlle Vertenoeil,Gaëlle Vertenoeil,Anna Ngo,Ann Koay,Hana Raslova,Pierre J. Courtoy,Meng Ling Choong,Isabelle Plo,William Vainchenker,Robert Kralovics,Stefan N. Constantinescu,Stefan N. Constantinescu +24 more
TL;DR: This study provides a novel signaling paradigm, whereby a mutated chaperone constitutively activates cytokine receptor signaling, and knocking down either MPL/TpoR or JAK2 in megakaryocytic progenitors from patients carrying CALR mutations inhibited cytokine-independent megakARYocytic colony formation.
Journal ArticleDOI
Calreticulin mutants in mice induce an MPL-dependent thrombocytosis with frequent progression to myelofibrosis.

Constantinescu,Stefan N. Constantinescu,Isabelle Plo,Isabelle Plo,Isabelle Plo,William Vainchenker,William Vainchenker,William Vainchenker +28 more
TL;DR: It is confirmed in c-mpl- and tpo-deficient mice that expression of Mpl, but not of Tpo, was essential for the CALR mutants to induce thrombocytosis in vivo, although Tpo contributes to disease penetrance.
Journal ArticleDOI
Myeloproliferative neoplasm induced by constitutive expression of JAK2V617F in knock-in mice.
Caroline Marty,Catherine Lacout,Catherine Lacout,Catherine Lacout,Antoine Martin,Salma Hasan,Salma Hasan,Salma Hasan,Sylvie Jacquot,Marie-Christine Birling,William Vainchenker,William Vainchenker,William Vainchenker,Jean-Luc Villeval,Jean-Luc Villeval,Jean-Luc Villeval +15 more
TL;DR: Investigating the effect of an endogenous heterozygous expression of JAK2(V617F) in knock-in (KI) mice found that it results in severe PV-like disease with secondary myelofibrosis and not in ET- like disease as expected from patient study.
Journal ArticleDOI
A role for reactive oxygen species in JAK2 V617F myeloproliferative neoplasm progression.
Caroline Marty,Catherine Lacout,Catherine Lacout,Nathalie Droin,Nathalie Droin,J-P Le Couedic,J-P Le Couedic,Vincent Ribrag,Vincent Ribrag,Eric Solary,Eric Solary,William Vainchenker,William Vainchenker,J-L Villeval,J-L Villeval,Isabelle Plo,Isabelle Plo +16 more
TL;DR: It is shown that JAK2V617F induces the accumulation of reactive oxygen species (ROS) in the hematopoietic stem cell compartment of a knock-in (KI) mouse model and in patients with JAK1 V617F MPNs, and targeting ROS accumulation might prevent the development of JAK 2V617f MPNs.
