TL;DR: It is concluded that SkQs are rechargeable, mitochondria-targeted antioxidants of very high efficiency and specificity and might be used to effectively prevent ROS-induced oxidation of lipids and proteins in the inner mitochondrial membrane in vivo.
Journal ArticleDOI
“Wages of Fear”: transient threefold decrease in intracellular ATP level imposes apoptosis
D. S. Izyumov,A. V. Avetisyan,Olga Yu. Pletjushkina,Dmitrii V. Sakharov,Karel W. A. Wirtz,Boris V. Chernyak,Vladimir P. Skulachev +6 more
TL;DR: The cell suicide is resistant to cyclosporine A, a phospholipase inhibitor trifluoroperazine, the JNK and p38 kinase inhibitors, oligomycin, N-acetyl cysteine and mitoQ, differing in these respects from the tumor necrosis factor- and H(2)O(2)-induced apoptoses.
Journal ArticleDOI
Oligomycin, inhibitor of the F0 part of H+-ATP-synthase, suppresses the TNF-induced apoptosis.
Liarisa A. Shchepina,Olga Yu. Pletjushkina,A. V. Avetisyan,Liora E. Bakeeva,Fetisova Ek,D. S. Izyumov,V. B. Saprunova,Mikhail Yu. Vyssokikh,Boris V. Chernyak,Vladimir P. Skulachev +9 more
TL;DR: It is shown that release of cytochrome c and apoptosis induced by tumor necrosis factor α (TNF) in HeLa cells can be inhibited by (i) overexpression of an oncoprotein Bcl-2, (ii) Cyclosporin A, an inhibitor of the mitochondrial permeability transition pore (PTP) or (iii) oligomycin, an inhibitors of H+- ATP-synthase.
Journal ArticleDOI
Production of reactive oxygen species in mitochondria of HeLa cells under oxidative stress.
Boris V. Chernyak,D. S. Izyumov,Konstantin G. Lyamzaev,A. A. Pashkovskaya,Olga Yu. Pletjushkina,Yuri N. Antonenko,Dmitrii V. Sakharov,Karel W. A. Wirtz,Vladimir P. Skulachev +8 more
TL;DR: It is concluded that the initial segments of the respiratory chain can be an important source of ROS, which are targeted to mitochondria, determining the fate of the cell subjected to oxidative stress.
Journal ArticleDOI
Prevention of cardiolipin oxidation and fatty acid cycling as two antioxidant mechanisms of cationic derivatives of plastoquinone (SkQs).
Vladimir P. Skulachev,Yury N. Antonenko,Dmitry A. Cherepanov,Boris V. Chernyak,D. S. Izyumov,Ludmila S. Khailova,S.S. Klishin,Galina A. Korshunova,Konstantin G. Lyamzaev,Olga Yu. TL;DR: In mitochondria, the cycling gives rise to mild uncoupling, thereby decreasing membrane potential and ROS generation coupled to reverse electron transport in the respiratory chain and in yeast cells, dodecyltriphenylphosphonium (capital ES, Cyrillic12TPP), the cationic part of SkQ1, induces uncoupled that is mitochondria-targeted.
