Journal ArticleDOI
Understanding Asthma Phenotypes, Endotypes, and Mechanisms of Disease.
TL;DR: The molecular mechanisms behind the heterogeneity of airway inflammation in asthmatic patients are presented, which have shifted existing paradigms in the approach to asthma to tailor novel therapies.
Journal ArticleDOI
Distinct Effector B Cells Induced by Unregulated Toll-like Receptor 7 Contribute to Pathogenic Responses in Systemic Lupus Erythematosus
Scott A. Jenks,Kevin S. Cashman,Esther Zumaquero,Urko M. Marigorta,Aakash V. Patel,Xiaoqian Wang,Deepak Tomar,Matthew C. Woodruff,Zoe Simon,Regina Bugrovsky,Emily L. Blalock,Christopher D. Scharer,Christopher M. Tipton,Chungwen Wei,S. Sam Lim,Michelle Petri,Timothy B. Niewold,Jennifer H. Anolik,Greg Gibson,F. Eun-Hyung Lee,Jeremy M. Boss,Frances E. Lund,Ignacio Sanz +22 more
TL;DR: A distinct differentiation fate of autoreactive naive B cells into PC precursors with hyper‐responsiveness to innate stimuli is defined as well as establishes prominence of extra‐follicular B cell activation in SLE, and identifies therapeutic targets.
Journal ArticleDOI
Extrafollicular B cell responses correlate with neutralizing antibodies and morbidity in COVID-19.
Matthew C. Woodruff,Richard P. Ramonell,Doan C. Nguyen,Kevin S. Cashman,Ankur Singh Saini,Natalie S. Haddad,Ariel Ley,Shuya Kyu,J. Christina Howell,Tugba Ozturk,Saeyun Lee,Naveenchandra Suryadevara,James Brett Case,Regina Bugrovsky,Weirong Chen,Jacob Estrada,Andrea Morrison-Porter,Andrew Derrico,Fabliha Anam,Monika Sharma,Henry M. Eun-Hyung Lee,Ignacio Sanz +32 more
TL;DR: Overall, these findings strongly suggest a pathogenic role for immune activation in subsets of patients with COVID-19, and provide further evidence that targeted immunomodulatory therapy may be beneficial in specific patient subpopulations and can be informed by careful immune profiling.
Journal ArticleDOI
Rapid isolation and profiling of a diverse panel of human monoclonal antibodies targeting the SARS-CoV-2 spike protein.
Seth J. Zost,Pavlo Gilchuk,Rita E. Chen,James Brett Case,Joseph X. Reidy,Andrew Trivette,Rachel S. Nargi,Rachel E. Sutton,Naveenchandra Suryadevara,Elaine C. TL;DR: A platform for rapid antibody discovery enabled the isolation of hundreds of human monoclonal antibodies against SARS-CoV-2 and the prioritization of potent antibody candidates for clinical trials in patients with COVID-19, and demonstrates the speed and robustness of advanced antibody discovery platforms.
Journal ArticleDOI
Diversity, cellular origin and autoreactivity of antibody-secreting cell population expansions in acute systemic lupus erythematosus
Christopher M. Tipton,Christopher F. Fucile,Jaime Darce,Asiya Seema Chida,Travis Ichikawa,Ivan V. Gregoretti,Sandra M Schieferl,Jennifer Hom,Scott A. Jenks,Ron J. Feldman,Ramit Mehr,Chungwen Wei,F. Eun-Hyung Lee,Wan Cheung Cheung,Alexander F. Rosenberg,Iñaki Sanz +15 more
TL;DR: Deep sequencing, proteomic profiling of autoantibodies and single-cell analysis demonstrated highly diversified ASCs punctuated by clones expressing the variable heavy-chain region VH4-34 that produced dominant serum autoantIBodies, shed light on the pathogenesis of SLE and explain the benefit of agents that target B cells and should facilitate the design of future therapies.
