Size-dependent toxicity and cell interaction mechanisms of gold nanoparticles on mouse fibroblasts
Rosella Coradeghini,Sabrina Gioria,César Pascual García,Paola Nativo,Fabio Franchini,Douglas Gilliland,Jessica Ponti,François Rossi +7 more
TL;DR: The mechanism of NPs cell internalization was investigated using immunocytochemistry and western blot, and no significant effect was observed in the expression level of caveolin, while reduction of the expression and degradation of the clathrin heavy chain was observation in cells exposed for 72 h to AuNPs.
Journal ArticleDOI
Protein−Nanoparticle Interaction: Identification of the Ubiquitin−Gold Nanoparticle Interaction Site
TL;DR: It is demonstrated that it is possible to identify the protein--nanoparticle interaction site at amino acid scale in solution using NMR, chemical shift perturbation analysis, and dynamic light scattering.
Journal ArticleDOI
Comprehensive In Vitro Toxicity Testing of a Panel of Representative Oxide Nanomaterials: First Steps towards an Intelligent Testing Strategy
Lucian Farcal,Fernando Torres Andón,Luisana Di Cristo,Bianca Maria Rotoli,Ovidio Bussolati,Enrico Bergamaschi,Agnieszka Mech,Nanna B. Hartmann,Kirsten Rasmussen,Juan Riego-Sintes,Jessica Ponti,Agnieszka Kinsner-Ovaskainen,François Rossi,Agnes G. Oomen,Peter A. Bos,Rui Chen,Ru Bai,Chunying Chen,Louise Rocks,Norma Fulton,B. Ross,Gary R Hutchison,Lang Tran,Sarah Mues,Rainer Ossig,Jürgen Schnekenburger,Luisa Campagnolo,Lucia Vecchione,Antonio Pietroiusti,Bengt Fadeel +29 more
TL;DR: Overall, this testing approach, based on a battery of cellular systems and test assays, complemented by an exhaustive physico-chemical characterization of NMs, could be deployed for the development of an ITS suitable for risk assessment of N Ms.
Journal ArticleDOI
Problems and challenges in the development and validation of human cell-based assays to determine nanoparticle-induced immunomodulatory effects
Gertie Janneke Oostingh,Eudald Casals,Paola Italiani,Renato Colognato,René Stritzinger,Jessica Ponti,Tobias Pfaller,Yvonne Kohl,Daniëlla Ooms,Flavia Favilli,H. Leppens,Davide Lucchesi,François Rossi,Inge Nelissen,Hagen Thielecke,Víctor F. Puntes,Albert Duschl,Diana Boraschi +17 more
TL;DR: The problems that were encountered in the immunological assay systems used in this study, such as chemical or endotoxin contamination and optical interference caused by the dense material, significantly affected the data obtained.
Journal ArticleDOI
Gold nanoparticles downregulate interleukin-1β-induced pro-inflammatory responses.
Vadim V. Sumbayev,Inna M. Yasinska,César Pascual García,Douglas Gilliland,Gurprit S. Lall,Bernhard F. Gibbs,David R. Bonsall,Luca Varani,François Rossi,Luigi Calzolai +9 more
TL;DR: It is shown for the first time that citrate-stabilized gold nanoparticles, in a size dependent manner, specifically downregulate cellular responses induced by IL-1β both in vitro and in vivo.
