Journal ArticleDOI
Ionic immune suppression within the tumour microenvironment limits T cell effector function
Robert L. Eil,Suman K. Vodnala,David Clever,Christopher A. Klebanoff,Christopher A. Klebanoff,Madhusudhanan Sukumar,Jenny H. Pan,Douglas C. Palmer,Alena Gros,Tori N. Yamamoto,Shashank J. Patel,Geoffrey Guittard,Zhiya Yu,Valentina Carbonaro,Klaus Okkenhaug,David S. Schrump,W. Marston Linehan,Rahul Roychoudhuri,Nicholas P. Restifo +18 more
TL;DR: An ionic checkpoint that blocks T cell function in tumours is uncovered and potential new strategies for cancer immunotherapy are identified, including boosting potassium efflux in tumour-specific T cells by overexpressing the potassium channel Kv1.3.
Journal ArticleDOI
Cish actively silences TCR signaling in CD8+ T cells to maintain tumor tolerance
Douglas C. Palmer,Geoffrey Guittard,Zulmarie Franco,Joseph G. Crompton,Robert L. Eil,Shashank J. Patel,Yun Ji,Nicholas J. Van Panhuys,Christopher A. Klebanoff,Madhusudhanan Sukumar,David Clever,David Clever,Anna Chichura,Rahul Roychoudhuri,Rajat Varma,Ena Wang,Luca Gattinoni,Francesco M. Marincola,Lakshmi Balagopalan,Lawrence E. Samelson,Nicholas P. Restifo +20 more
TL;DR: It is found that Cish, a member of the SOCS family, is induced by TCR stimulation in CD8+ T cells and inhibits their functional avidity against tumor.
Journal ArticleDOI
Cutting edge: Dok-1 and Dok-2 adaptor molecules are regulated by phosphatidylinositol 5-phosphate production in T cells.
Geoffrey Guittard,Audrey Gérard,Sophie Dupuis-Coronas,Hélène Tronchère,Eva Mortier,Cédric Favre,Daniel Olive,Pascale Zimmermann,Bernard Payrastre,Jacques A. Nunès +9 more
TL;DR: This work finds that Dok-1/Dok-2 PH domains bind in vitro to the rare phosphoinositide species, phosphatidylinositol 5-phosphate (PtdIns5P) and indicates that PH-Ptdins5P interactions regulate T cell responses.
Journal ArticleDOI
B7-DC-Ig enhances vaccine effect by a novel mechanism dependent on PD-1 expression level on T cell subsets
Mikayel Mkrtichyan,Yana G. Najjar,Estella C. Raulfs,Linda Liu,Solomon Langerman,Geoffrey Guittard,Laurent Ozbun,Samir N. Khleif +7 more
TL;DR: It is demonstrated that B7-DC-Ig can enhance the therapeutic efficacy of vaccine when combined with cyclophosphamide and this combination significantly enhances Ag-specific immune responses and leads to complete eradication of established tumors in 60% of mice and that this effect is CD8 dependent.
Journal ArticleDOI
Vitamin D Controls Tumor Growth and CD8+ T Cell Infiltration in Breast Cancer.
Esma Karkeni,Stéphanie O. Morin,Berna Bou Tayeh,Armelle Goubard,Emmanuelle Josselin,Rémy Castellano,Cyril Fauriat,Geoffrey Guittard,Daniel Olive,Jacques A. Nunès +9 more
TL;DR: It is shown that vitamin D decreases breast cancer tumor growth and inflammation in the tumor microenvironment in vivo, and it is believed that supplementation with vitamin D can in certain conditions represent a new adjuvant in the treatment of breast cancers.
