Journal ArticleDOI
Subcellular localization of proteasomes and their regulatory complexes in mammalian cells
Paul Brooks,Graciela Fuertes,Rachael Z. Murray,Suchira Bose,Erwin Knecht,Martin Rechsteiner,Klavs B. Hendil,Keiji Tanaka,Julian Dyson,A.Jennifer Rivett +9 more
TL;DR: In this article, the subcellular distribution of the different forms of proteasomes using subunit specific antibodies is investigated, and it is shown that different regulatory complexes and subpopulations have different distributions within mammalian cells and, therefore, that the distribution is more complex than has been reported for yeast proteasome.
Journal ArticleDOI
Disturbed Cholesterol Traffic but Normal Proteolytic Function in LAMP-1/LAMP-2 Double-deficient Fibroblasts
Eeva-Liisa Eskelinen,Christine K. Schmidt,Silja Neu,Marion Willenborg,Graciela Fuertes,Natalia Salvador,Yoshitaka Tanaka,Renate Lüllmann-Rauch,Dieter Hartmann,Jörg Heeren,Kurt von Figura,Erwin Knecht,Paul Saftig +12 more
TL;DR: Findings indicate partially overlapping functions for LAMP-1 and -2 in lysosome biogenesis, autophagy, and cholesterol homeostasis as well as reduced amounts of lipid droplets in double deficient mice.
Journal ArticleDOI
Subpopulations of proteasomes in rat liver nuclei, microsomes and cytosol
A. Palmer,A J Rivett,Stuart Thomson,Klavs B. Hendil,Geoffrey W. Butcher,Graciela Fuertes,Erwin Knecht +6 more
TL;DR: Although there appear not to be differences in proteasome composition in the alpha subunits (C8 and C9) in the different locations, the relative amounts of some beta subunits varied, which probably reflect differences in the function of proteasomes in distinct cell compartments.
Journal ArticleDOI
Changes in the proteolytic activities of proteasomes and lysosomes in human fibroblasts produced by serum withdrawal, amino-acid deprivation and confluent conditions.
Graciela Fuertes,José Javier Martín de Llano,Adoración Villarroya,A. Jennifer Rivett,Erwin Knecht +4 more
TL;DR: The results show that the activity of the various proteolytic pathways depends on the growth conditions of cells and will be useful for investigation of the specific signals that control their activity.
Journal ArticleDOI
Role of proteasomes in the degradation of short-lived proteins in human fibroblasts under various growth conditions.
TL;DR: It is concluded that, although proteasomes are responsible of about 40-60% of the degradation of short-lived proteins in normal human fibroblasts, lysosomes have also an important participation in the degradation with a half-life of approximately 1h.
