Secretory Mechanisms and Intercellular Transfer of MicroRNAs in Living Cells
Nobuyoshi Kosaka,Haruhisa Iguchi,Yusuke Yoshioka,Fumitaka Takeshita,Yasushi Matsuki,Takahiro Ochiya +5 more
TL;DR: It is shown that miRNAs are released through a ceramide-dependent secretory machinery and that the secretory miRNAAs are transferable and functional in the recipient cells and that a tumor-suppressive miRNA secreted via this pathway was transported between cells and exerted gene silencing in the recipients cells, thereby leading to cell growth inhibition.
Journal ArticleDOI
Circulating microRNA in body fluid: a new potential biomarker for cancer diagnosis and prognosis.
TL;DR: The usefulness of circulating miRNA for cancer diagnosis, prognosis, and therapeutics is summarized and a mechanism for the secretion and incorporation of miRNA into the cells is proposed.
Journal ArticleDOI
Activation of peroxisome proliferator-activated receptor δ induces fatty acid β-oxidation in skeletal muscle and attenuates metabolic syndrome
Toshiya Tanaka,Joji Yamamoto,Satoshi Iwasaki,Hiroshi Asaba,Hiroki Hamura,Yukio Ikeda,Mitsuhiro Watanabe,Kenta Magoori,Ryoichi X. Ioka,Keisuke Tachibana,Yuichiro Watanabe,Yasutoshi Uchiyama,Koichi Sumi,Haruhisa Iguchi,Sadayoshi Ito,Takefumi Doi,Takao Hamakubo,Makoto Naito,Johan Auwerx,Masashi Yanagisawa,Tatsuhiko Kodama,Juro Sakai +21 more
TL;DR: Analysis of rat L6 myotubes treated with the PPARδ subtype-selective agonist, GW501516, by the Affymetrix oligonucleotide microarrays revealed that PPAR Δ is pivotal to control the program for fatty acid oxidation in the skeletal muscle, thereby ameliorating obesity and insulin resistance through its activation in obese animals.
Journal ArticleDOI
Neutral sphingomyelinase 2 (nSMase2)-dependent exosomal transfer of angiogenic microRNAs regulate cancer cell metastasis.
Nobuyoshi Kosaka,Haruhisa Iguchi,Keitaro Hagiwara,Yusuke Yoshioka,Fumitaka Takeshita,Takahiro Ochiya +5 more
TL;DR: It is demonstrated that the neutral sphyngomyelinase 2 (nSMase2) regulates exosomal microRNA (miRNA) secretion and promotes angiogenesis within the tumor microenvironment as well as metastasis, suggesting that the horizontal transfer of exosome miRNAs from cancer cells can dictate the microenviromental niche for the benefit of the cancer cell.
Journal ArticleDOI
Role of STAT-3 in regulation of hepatic gluconeogenic genes and carbohydrate metabolism in vivo
Hiroshi Inoue,Wataru Ogawa,Michitaka Ozaki,Sanae Haga,Michihiro Matsumoto,Kensuke Furukawa,Naoko Hashimoto,Yoshiaki Kido,Toshiyuki Mori,Hiroshi Sakaue,Kiyoshi Teshigawara,Shiyu Jin,Haruhisa Iguchi,Ryuji Hiramatsu,Derek LeRoith,Kiyoshi Takeda,Shizuo Akira,Masato Kasuga +17 more
TL;DR: Liver-specific expression of a constitutively active form of STAT-3, achieved by infection with an adenovirus vector, markedly reduced blood glucose, plasma insulin concentrations and hepatic gluconeogenic gene expression in diabetic mice.
