Journal ArticleDOI
Structural basis of Nav1.7 inhibition by an isoform-selective small-molecule antagonist.
Shivani Ahuja,Susmith Mukund,Lunbin Deng,Kuldip Khakh,Elaine Chang,Hoangdung Ho,Stephanie Shriver,Clint Young,Sophia Lin,J. P. Johnson,Ping Wu,Jun Li,Mary Coons,Christine Tam,Bobby Brillantes,Honorio Sampang,Kyle Mortara,Krista K. Bowman,Kevin R Clark,Alberto Estevez,Zhiwei Xie,Henry Verschoof,Michael Edward Grimwood,Christoph Martin Dehnhardt,Jean-Christophe Andrez,Thilo Focken,Daniel P. Sutherlin,Brian Safina,Melissa A. Starovasnik,Daniel F. Ortwine,Yvonne Franke,Charles J. Cohen,David H. Hackos,Christopher M. Koth,Jian Payandeh +34 more
TL;DR: A novel protein-engineering strategy is devised that overcomes the technical complexities of producing full-length human Nav channels and demonstrates how this important new class of gating modifier engages VSD4 to inhibit Nav channel activity through a “voltage-sensor trapping” mechanism.
Journal ArticleDOI
An Unbiased Screen for Human Cytomegalovirus Identifies Neuropilin-2 as a Central Viral Receptor.
Nadia Martinez-Martin,Jessica Marcandalli,Christine S. Huang,Christopher P. Arthur,Michela Perotti,Mathilde Foglierini,Hoangdung Ho,Annie M. Dosey,Stephanie Shriver,Jian Payandeh,Alexander Leitner,Antonio Lanzavecchia,Antonio Lanzavecchia,Laurent Perez,Claudio Ciferri +14 more
TL;DR: A library consisting of most single transmembrane human receptors is built and a workflow for unbiased and high-sensitivity detection of receptor-ligand interactions is implemented to elucidate the long-sought receptor of human cytomegalovirus (HCMV), the leading viral cause of congenital birth defects.
Journal ArticleDOI
Structural basis for dual-mode inhibition of the ABC transporter MsbA
Hoangdung Ho,Anh Miu,Mary Kate Alexander,Natalie K. Garcia,Angela Oh,Inna Zilberleyb,Mike Reichelt,Cary D. Austin,Christine Tam,Stephanie Shriver,Huiyong Hu,Sharada Labadie,Jun Liang,Lan Wang,Jian Wang,Yan Lu,Hans E. Purkey,John G. Quinn,Yvonne Franke,Kevin R Clark,Maureen Beresini,Man-Wah Tan,Benjamin D. Sellers,Till Maurer,Michael F. T. Koehler,Aaron T. Wecksler,James R. Kiefer,Vishal Verma,Yiming Xu,Mireille Nishiyama,Jian Payandeh,Christopher M. Koth +31 more
TL;DR: A framework for the selective modulation of ABC transporters is established and provides rational avenues for the design of new antibiotics and other therapeutics targeting this protein family.
Journal ArticleDOI
The Structural Basis for Class II Cytokine Receptor Recognition by JAK1
Ryan Ferrao,Heidi J.A. Wallweber,Hoangdung Ho,Christine Tam,Yvonne Franke,John G. Quinn,Patrick J. Lupardus +6 more
TL;DR: Two crystal structures of the human JAK1 FERM and SH2 domains bound to peptides derived from the class II cytokine receptors IFN-λ receptor 1 and IL-10 receptor 1 (IFNLR1 and IL10RA) are presented to identify a conserved JAK binding site for receptor peptides and elucidate the mechanism by which class II inflammation receptors interact with JAK 1.
Journal ArticleDOI
Disrupting Gram-Negative Bacterial Outer Membrane Biosynthesis through Inhibition of the Lipopolysaccharide Transporter MsbA
Mary Kate Alexander,Anh Miu,Angela Oh,Mike Reichelt,Hoangdung Ho,Cecile Chalouni,Sharada Labadie,Lan Wang,Jun Liang,Nicholas N. Nickerson,Huiyong Hu,Lan Yu,Miaofen Du,Donghong Yan,Summer Park,Janice Kim,Min Xu,Benjamin D. Sellers,Hans E. Purkey,Nicholas J. Skelton,Michael F. T. Koehler,Jian Payandeh,Vishal Verma,Yiming Xu,Christopher M. Koth,Mireille Nishiyama +25 more
TL;DR: A biochemical screen for inhibitors of MsbA was conducted and a series of quinoline compounds that kill Escherichia coli through inhibition of its ATPase and transport activity were identified, with no loss of activity against clinical multidrug-resistant strains.
