MLL targets SET domain methyltransferase activity to Hox gene promoters.
Thomas A. Milne,Scott D. Briggs,Hugh W. Brock,Mary Ellen Martin,Denise Gibbs,C. David Allis,Jay L. Hess +6 more
TL;DR: It is shown that MLL regulates target Hox gene expression through direct binding to promoter sequences and the MLL SET domain is a histone H3 lysine 4-specific methyltransferase whose activity is stimulated with acetylated H3 peptides.
Journal ArticleDOI
Altered Hox expression and segmental identity in Mll -mutant mice
TL;DR: Mll is required for proper segment identity in mammals, displays haplo-insufficiency, and positively regulates Hox gene expression.
Journal ArticleDOI
Physical Association and Coordinate Function of the H3 K4 Methyltransferase MLL1 and the H4 K16 Acetyltransferase MOF
Yali Dou,Thomas A. Milne,Thomas A. Milne,Alan J. Tackett,Edwin R. Smith,Aya Fukuda,Joanna Wysocka,C. David Allis,Brian T. Chait,Jay L. Hess,Robert G. Roeder +10 more
TL;DR: An activator-based mechanism for joint MLL1 and MOF recruitment and targetedmethylation and acetylation is indicated and a molecular explanation for the closely correlated distribution of H3 K4 methylation and H4 K16 acetylations on active genes is provided.
Journal ArticleDOI
Menin Associates with a Trithorax Family Histone Methyltransferase Complex and with the Hoxc8 Locus
Christina M. Hughes,Orit Rozenblatt-Rosen,Thomas A. Milne,Terry D. Copeland,Stuart S. Levine,Jeffrey C. Lee,D. Neil Hayes,Kalai Selvi Shanmugam,Arindam Bhattacharjee,Christine A. Biondi,Graham F. Kay,Nicholas K. Hayward,Jay L. Hess,Matthew Meyerson +13 more
TL;DR: It is shown that menin is associated with a histone methyltransferase complex containing two trithorax family proteins, MLL2 and Ash2L, and other homologs of the yeast Set1 assembly, and that this activity is related to tumor suppression by MEN1.
Journal ArticleDOI
Menin and MLL cooperatively regulate expression of cyclin-dependent kinase inhibitors
Thomas A. Milne,Christina M. Hughes,Ricardo V. Lloyd,Zhaohai Yang,Orit Rozenblatt-Rosen,Yali Dou,Robert W. Schnepp,Cynthia Krankel,Virginia A. LiVolsi,Denise Gibbs,Xianxin Hua,Robert G. Roeder,Matthew Meyerson,Jay L. Hess +13 more
TL;DR: It is shown that menin directly regulates expression of the cyclin-dependent kinase inhibitors p27Kip1 and p18Ink4c and plays a central role in menin's activity as a tumor suppressor.
