A novel Epac-specific cAMP analogue demonstrates independent regulation of Rap1 and ERK.
Jorrit M. Enserink,Anne Elisabeth Christensen,Johan de Rooij,Miranda van Triest,Frank Schwede,Hans G. Genieser,Stein Ove Døskeland,Jonathan L. Blank,Johannes L. Bos +8 more
TL;DR: A novel cAMP analogue is developed, 8CPT-2Me-cAMP, which activates Epac, but not PKA, both in vitro and in vivo, and tests the widespread model that Rap1 mediates cAMP-induced regulation of the extracellular signal-regulated kinase (ERK).
Journal ArticleDOI
cAMP Analog Mapping of Epac1 and cAMP Kinase DISCRIMINATING ANALOGS DEMONSTRATE THAT Epac AND cAMP KINASE ACT SYNERGISTICALLY TO PROMOTE PC-12 CELL NEURITE EXTENSION
Anne Elisabeth Christensen,Frode Selheim,Johan de Rooij,Sarah Dremier,Frank Schwede,Khanh K. Dao,Aurora Martinez,Carine Maenhaut,Johannes L. Bos,Hans G. Genieser,Stein Ove Døskeland +10 more
TL;DR: In this paper, the relative role of cAMP-dependent protein kinase (cAPK) and guanine exchange factor directly activated by cAMP (Epac) was investigated.
Journal ArticleDOI
Cyclic AMP induces integrin-mediated cell adhesion through Epac and Rap1 upon stimulation of the β2-adrenergic receptor
Savithri Rangarajan,Jorrit M. Enserink,H. Bea Kuiperij,Johan de Rooij,Leo S. Price,Frank Schwede,Johannes L. Bos +6 more
TL;DR: It is demonstrated that external stimuli of cAMP signaling, i.e., isoproterenol, which activates the Gαs-coupled β2-adrenergic receptor can induce integrin-mediated cell adhesion through the Epac-Rap1 pathway.
Journal ArticleDOI
α-catenin cytomechanics--role in cadherin-dependent adhesion and mechanotransduction
Adrienne K. Barry,Hamid Tabdili,Ismaeel Muhamed,Jun Wu,Nitesh Shashikanth,Guillermo A. Gomez,Alpha S. Yap,Cara J. Gottardi,Johan de Rooij,Ning Wang,Deborah E. Leckband +10 more
TL;DR: Bead-twisting measurements andplementary investigations of cadherin-based rigidity sensing suggest that, although &agr;-catenin alters traction force generation, it is not the sole regulator of cell contractility on compliant cadhersin-coated substrata.
Journal ArticleDOI
Protein kinase B activation and lamellipodium formation are independent phosphoinositide 3-kinase-mediated events differentially regulated by endogenous Ras.
David H.J. van Weering,Johan de Rooij,Barbara M. Marte,Julian Downward,Johannes L. Bos,Boudewijn M.T. Burgering +5 more
TL;DR: It is concluded that, in vivo, p85 and Ras synergize to activate PI 3-kinase and that strong activation of only endogenous Ras exerts a small effect on PI 3 -kinase activity, sufficient for PKB activation but not lamellipodium formation.
